千金苇茎汤含药血清对香烟烟雾暴露小鼠脾Naive T细胞Th17/Treg分化及Notch通路表达的影响

摘　　要：目的旨在实验观察千金苇茎汤(Phragmitis Decoction,PD)大鼠含药血清对香烟烟雾水溶液处理的小鼠脾Naive T细胞分化Th17/Treg平衡及Notch信号通路表达的影响。方法将雄性健康SD大鼠30只随机分为A和B组(生理盐水5 ml/kg·bw)、C组(100 mg/kg·bw乙酰半胱氨酸)、D组(10 g/kg·bw PD血清)、E组(5 g/kg·bw PD血清)、F组(2.5 g/kg·bw PD血清),每日灌胃2次,连续3 d。末次给药1 h后,股动脉取血制备含药血清,高效液相色谱法(HPLC)含药血清浓度。用免疫磁珠分选C57BL/6雌性健康小鼠脾Naive T细胞,通过细胞培养后将其分为6组,分别标记为空白组(接受体积无药血清DMEM培养)、模型组(接受5.0%香烟烟雾水溶液的DMEM培养)、西药血清干预组(接受含乙酰半胱氨酸血清培养)、PD高剂量血清干预组(接受含高剂量PD血清培养)、千金苇茎汤中剂量血清干预组(接受含中剂量PD血清培养)、PD低剂量血清干预组(接受含低剂量PD血清培养)。流式细胞技术检测各组Naive T细胞Th17/Treg数量及比率变化。Western blot检测Naive T细胞Notch、Jagged-1、Jagged-2、Delta-like-1、Delta-like-3和Delta-like-4等蛋白表达。采用ELISA法检测各组中Naive T细胞培养液IL-17、IL-6、IL-10和TGF-β的含量。结果PD高剂量血清干预组及西药血清干预组Th17细胞相对数量明显低于模型组(P<0.05),PD高剂量血清干预组Th17细胞相对数量明显低于西药组(P<0.05)。PD高和中剂量血清干预组及血清西药干预组Treg细胞相对数量明显高于模型组(P<0.05)。PD高剂量血清干预组Delta-like-1、Delta-like-3和Delta-like-4表达明显低于模型组(P<0.05),PD高剂量血清干预组Jagged-1和Jagged-2的表达水平明显高于模型组(P<0.05),也明显高于西药血清干预组(P<0.05)。PD高和中剂量血清干预组及西药血清干预组的IL-17表达明显低于模型组(P<0.05);PD高和中剂量血清干预组IL-17表达明显低于西药血清干预组(P<0.05)。PD高剂量Objective To investigate the effects of phragmitis decoction(PD)-containing serum on Th17/Treg balance and Notch signaling pathway expression of spleen Naive T cells of mice dealed by water-soluble concents of cigarette smoke.Methods Thirty healthy male SD rats were randomly divided into group A and group B(normal saline at 5 ml/kg·bw),group C(acetylcysteine at 100 mg/kg·bw),group D(PD serum at 10 g/kg·bw),group E(PD serum at 5 g/kg·bw)and group F(PD serum at 2.5 g/kg·bw).The rats were intragastrically administered twice a day for three consecutive days.One hour after the last administration,blood was taken from femoral artery to prepare serum containing the drug,and the concentration of serum containing the drug was determined by HPLC.The spleen Naive T cells of C57 BL/6 female healthy mice were sorted by immunomagnetic beads,then these cells were divided into blank group(DMEM culture with drug-free serum),model group(DMEM culture with 5.0%aqueous solution of cigarette smoke),western medicine serum intervention group(DMEM culture with acetylcysteine),PD high-dose serum intervention group(DMEM culture with high-dose serum),PD medium-dose serum intervention group(DMEM culture with medium-dose serum)and PD low-dose serum intervention group(DMEM culture with low-dose serum).Flow cytometry was used to detect the number and ratio of Th17/Treg in each group.Western blot was used to detect the expressions of notch,Jagged-1,Jagged-2,Delta-like-1,delta-like-3 and delta-like-4 in Naive T cells of each group.The levels of IL-17,IL-6,IL-10 and TGF-β of Naive T cells were detected by ELISA in each group.Results The relative number of Th17 cells in PD high-dose serum intervention group and western medicine serum intervention group was significantly lower than that in model group(P<0.05),and the relative number of Th17 cells in PD high-dose serum intervention group was significantly lower than that in western medicine group(P<0.05).The relative number of Treg cells in PD high and medium dose serum intervention groups and
