摘  要:目的探讨藏药唐古特白刺果提取物改善2型糖尿病大鼠认知功能的作用机制。方法选取30只SD大鼠,其中20只用于制备2型糖尿病脑病(DE)大鼠模型(制备方法:给予大鼠高糖高脂饲料结合STZ尾静脉注射法建立)组;剩余10只用于设置对照组(NG)。持续喂养12 w后,将20只DE大鼠分为模型组(DM)、治疗组(DM+NTB),各10只。4w内,两组分别以相同剂量的生理盐水、唐古特白刺果提取物(3.6g/kg·d)灌胃,保证其他实验条件一致。采用HE染色法和免疫组化法,观察各组大鼠海马组织形态学变化,检测晚期糖基化终产物受体(advanced glyeation end product receptor,RAGE)、核因子κB P65(nuclear factor-κB p65,NF-κB P65)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-1β(interleukin-1β,IL-1β)表达情况,对所得结果进行相关性分析。结果相较于NG组,DM组大鼠海马组织的RAGE、NF-κB P65、TNF-α、IL-1β均明显升高(P<0.05),且染色结果显示大鼠海马组织病理损伤严重。DM+NTB组虽存在一定的病理损伤,且蛋白表达均升高(P<0.05),但对比DM组有明显改善,唐古特白刺果提取物可抑制RAGE/NF-κB炎症信号通路。结论唐古特白刺果提取物可抑制2型糖尿病大鼠RAGE/NF-κB炎症信号通路,这可能是其改善2型糖尿病大鼠认知功能的关键机制。Objective To conduct a deeply research on the mechanism how the Tibetan medicine NTB improves cognitive function of type 2 rats with diabetic encephalopathy via inhibiting RAGE/NF-κB signaling pathway.Methods 30 SPF-grade SD male rats were randomly picked out and 20 of them were injected STZ through caudal vein combined with high sugar and fat diet to found the type 2 diabetic encephalopathy(DE)rat model,and the remaining 10 rats were categorized as the control group(NG).After continuous feeding for 12 weeks,20 DE rats were randomly di-vided into model group(DM)and NTB treatment group(DM+NTB),each with 10.4 weeks later,both were separately gavaged with the same dose of saline and the extract of NTB(3.6g/kg,once a day),other experimental conditions en-sured to be the same.Utilize HE staining and immunohistochemistry to observe morphological changes of hippocampal tissue and detect expression levels of the receptor for advanced glycation end product(RAGE)、nuclear factor-κB p65(NF-κB P65)、tumor necrosis factor-α(TNF-α)and interleukin-lβ(IL-1β),and make a correlation analysis for the results.Results The expression levels of RAGE,NF-κB P65,TNF-αand IL.-Iβapparently elevated in the DM group compared with the NG group(P<0.05),and the result of stained hippocampal tssue implicates that the patho-logical damage was more severe.There still exists certain pathological impairment in DM+NTB group,while the ex-pression levels of each protein mentioned above elevated to some extent,but they obviously improved compared with DM group.Conclusion The extract of NTB can inhibit RAGE/NF-κB inflammatory signaling pathway,which may ex-plain the mechanism of preventing and improving cognitive dysfunction in type 2 diabetic rats.
