摘 要:利用SSR标记技术和I2-KI染色法指导大豆杂交种的选育。针对大豆杂交种的育种特点,以课题组的不育系与恢复系为材料,用SSR分子标记技术,筛选出20对SSR标记,呈现出多态性,用20对SSR标记对不育系JLSXCMS1,恢复系中119-99及两者的杂交组合JLSXCMS1×中119-99(定名为优势豆-A-5)进行亲子鉴定研究,鉴定制种生产的杂交种是否为真杂交种;用I2-KI染色法对杂交组合JLSXCMS1×中119-99的F1育性表现进行研究,检验是否符合“三系”大豆杂交种育性要求,保证杂交种正常结实,保证杂交种达到大豆杂交种种子质量标准。结果表明,14对SSR标记对不育系JLSXCMS1,恢复系中119-99具有多态性,优势豆-A-5的带型在9对SSR标记位点均为亲本JLSXCMS1和中119-99的互补带型,9对SSR标记为优势豆-A-5的亲子鉴定标记;花期花粉育性用I2-KI染色法镜检,JLSXCMS1不育系通过多代回交育成,2010、2011、2012年镜检花粉平均败育率为99.9992%,成熟期植株表现全部不育。优势豆-A-5可育花粉占花粉总数为48.0%~61.4%,成熟期植株全部正常结实。在SSR标记及F1花粉育性鉴定技术指导下,育成杂交种优势豆-A-5,2019年通过山西省农作物品种审定委员会审定(晋审豆20190002)。The biotechnology of SSR marker and the I2-KI staining method are used to guide the breeding of hybrid soybeans.According to the characteristics of soybean hybrid breeding,the material was the maternal sterile line and the parental restorer line came from the research group of hybrid soybeans.Using SSR molecular marker technology,20 pairs of SSR markers were screened and showed polymorphism.The primer screening and seed purity identification of soybean hybrid variety JLSXCMS1×Zhong119-99(Youshidou-A-5)and its parents(The maternal sterile line is JLSXCMS1,and the parental restorer line is Zhong119-99)were studied by using 20 pairs of SSR marker technology.To identify the authenticity of hybrids produced by seed production.The F1 fertility of the hybrid combination(JLSXCMS1×Zhong119-99)was studied by I2-KI staining.Test whether it meets the fertility requirements of"three-line"soybean hybrid,ensure the normal fruiting of the hybrid,and the hybrid seeds meet the quality standards of soybean hybrid seeds.The results showed that 14 pairs of SSR markers were polymorphic in sterile lines JLSXCMS1 and restorer lines Zhong119-99.Youshidou-A-5 had parents(JLSXCMS1,Zhong119-99)with complementary band type in 9 pairs of SSR markers,9 pairs of SSR markers were used for paternity identification of Youshidou-A-5.The fertility of pollen at the flowering stage was examined by I2-KI staining.The sterile lines of JLSXCMS1 were bred through multiple generations backcross.The average abortion rate of pollen was 99.9992%by microscope in 2010,2011 and 2012.At maturity,all plants showed sterility,fertile pollen of Youshidou-A-5 accounted for 48.0%~61.4%of total pollen.All plants set normally at mature stage,Youshidou-A-5 was bred by the techniques of SSR markers and F1 pollen fertility identification,Youshidou-A-5 was approved by Shanxi Crop Variety Examination and Approval Committee in 2019(Authorized number:Jinshendou20190002).
