摘 要:猪流行性腹泻是猪场新生仔猪腹泻的主要病原之一。为确定云南省曲靖市某猪场新生仔猪出现呕吐、腹泻和高死亡率的病因,采集病死仔猪肠道、肠系膜淋巴结、脾脏等组织病料,处理后提取病原核酸,通过RT-PCR进行导致仔猪腹泻的猪流行性腹泻病毒(PEDV)、猪传染性胃肠炎病毒、猪轮状病毒、猪伪狂犬病病毒、猪瘟病毒、猪繁殖与呼吸综合征病毒等病原核酸检测。结果显示,仅PEDV核酸阳性,其他病原均为阴性。对检出的PEDV阳性样品N基因进行序列分析发现:阳性样品N基因与河北的T10-HB2018毒株核苷酸同源性最高,为99.7%;与疫苗株AJ1102和LW/L毒株核苷酸同源性为97.4%~97.5%,在同一个分支上,均属于基因Ⅱ群;与经典毒株CV777株核苷酸同源性略低,为95.7%。结果表明,引起该仔猪群腹泻的主要病因为PEDV感染,流行毒株未发生变异,当前疫苗对其防控有效。依据分析结果,该场采取PEDV疫苗紧急免疫,使疫情得到有效控制。本研究对于指导该地区猪群腹泻病防控具有参考意义。As porcine epidemic diarrhea virus(PEDV)is one of major pathogens leading to diarrhea in newborn piglets,in order to identify the causes of vomiting,diarrhea and high mortality of newborn piglets in a farm in Qujing City of Yunnan Province,tissue lesions including intestines,mesenteric lymph nodes and spleens of dead piglets were collected and extracted for pathogenic nucleic acids after sample treatment,followed by detection of PEDV,transmissible gastroenteritis virus(TGEV),rotavirus(RV),porcine pseudorabies virus(PRV)and classical swine fever virus(CSFV),porcine reproductive and respiratory syndrome virus(PRRSV)and other pathogens that led to the diarrhea by RT-PCR.The results showed that only PEDV nucleic acids were detected positive and all other pathogens were negative.By analysis on the sequence of N gene of PEDV positive samples,it was found that the N gene shared the nucleotide homology of 99.7%with T10-HB2018 strain isolated in Hebei Province,which was highest;97.4%-97.5%with vaccine strain AJ102 and LW/1 strain,all of which belonged to gene clusterⅡin the same branch;and95.7%with classical strain CV777,which was lowest.It was concluded that the diarrhea was mainly caused by PEDV infection,the available vaccines were effective for its prevention and control because no variation had occurred in the prevalent strain.According to the analysis results,emergency immunization with PEDV vaccines was adopted to effectively control the epidemic.Some reference was provided for the prevention and control of diarrhea in pigs in Yunnan Province.
