摘  要:本试验对东大、四平梅花鹿两个群体中所筛选出的8个产茸量相关SNP位点,运用竞争性等位基因特异性PCR分型技术(KASP),在7个梅花鹿群体的190头梅花鹿个体中进行验证。结果表明,7个位点具有多态性,其中SNP1427_255427_255、SNP307_1564、SNP123_7847和SNP2027_423为中度多态性位点,SNP614_2585、SNP1881_130和SNP596_3774为低度多态性位点,SNP219_9123是CC基因型的单态位点;最小基因等位基因频率(MAF)介于0.049~0.396之间,平均值为0.188;各位点的观测杂合度(Ho)介于0.09~0.426,平均值为0.265;期望杂合度(He)介于0.082~0.49之间,平均值为0.282;多态信息含量(PIC)介于0.078~0.369,平均值为0.232;有效等位基因数(Ne)介于1.2~2.8,平均值为1.9。7个多态性位点中,SNP614_2585、SNP1881_130和SNP596_3774位点偏离Hardy-Weinberg平衡。一般线性模型对多态性SNP位点与产茸量进行关联分析,发现所验证的7个位点对产茸量的影响均不显著,但SNP307_156、SNP614_258和SNP596_377位点存在高产基因型。本研究为产茸量相关SNP的实际应用提供理论依据。In this study,8 SNP loci related to the antler production were screened from Dongda and Siping sika deer populations.The Kompetitive allele specific PCR(KASP)was used to verify the SNP loci in 190 sika deer individuals from 7 sika deer groups.The results showed that 7 loci were polymorphic,including SNP1427255427255、SNP3071564、SNP1237847 and SNP2027423 are moderate polymorphisms,SNP6142585、SNP1881130、SNP5963774,SNP2199123 are single loci of CC genotypes;The minimum allele frequency(MAF)ranged from 0.049 to 0.396,with an average of 0.188;The observed heterozygosity(HO)ranged from 0.09 to 0.426,with an average of 0.265;The expected heterozygosity(He)ranged from 0.082 to 0.49,with an average of 0.282;And the polymorphism information content(PIC)ranged from 0.078 to 0.369,with an average of 0.232.The number of effective alleles(NE)ranged from 1.2 to 2.8,with an average of 1.9.Among the 7 polymorphic sites,SNP6142585、SNP1881130、SNP5963774 sites deviated from Hardy-Weinberg equilibrium.The results showed that the seven sites had no significant effect on the yield of antler.But SNP307156、SNP614258、SNP596377 locus has a high yield genotype.This study could provide a theoretical basis for the practical application of SNP related to the antler yield.
