摘  要:目的:探讨芝麻素对低氧条件下鼠嗜铬细胞瘤细胞(PC12)β淀粉样蛋白(Aβ)和β位淀粉样前体蛋白裂解酶-1(BACE-1)表达的影响。方法:PC12细胞随机分为正常氧组、低氧组、低氧加芝麻素0.5、5.0和50μmol/L浓度干预组,采用酶联免疫吸附法测定(ELISA)检测Aβ1-42水平,免疫印迹试验(Western blot)检测BACE-1蛋白表达。结果:低氧组Aβ1-42水平高于正常氧组(P<0.05),在0.5、5和50μmol/L浓度芝麻素干预后,低氧组Aβ水平降低(均P<0.05);低氧组BACE-1的蛋白表达高于正常氧组(P<0.05),在0.5、5和50μmol/L芝麻素干预后,低氧组BACE-1蛋白水平降低(均P<0.05)。结论:在低氧条件下,PC12细胞Aβ1-42、BACE-1蛋白表达升高,芝麻素降低低氧条件下升高的Aβ1-42,BACE-1表达。Objective:To investigate the effect of sesamin on expression ofβ-amyloid protein(Aβ)andβ-position amyloid precursor protein lyase-1(BACE-1)in PC12 cells under hypoxia conditions.Methods:PC12 cells were randomly divided into normoxia group,hypoxia group,hypoxia plus sesamin concentration intervention groups of 0.5,5 and 50μmol/L.Aβ1-42 level was detected by enzyme-linked immunosorbent assay(ELISA).The expression of BACE-1 protein was detected by Western blot.Results:The level of Aβ1-42 in the hypoxia group was higher than that in the normoxia group(P<0.05).After the intervention of sesamin at concentrations of 0.5,5 and 50μmol/L,the Aβlevel in the hypoxia group decreased(P<0.05).The protein expression of BACE-1 in the hypoxia group was higher than that of the normoxia group(P<0.05).After the intervention of 0.5,5 and 50μmol/L sesamin,the protein level of BACE-1 in the hypoxia gro up decreased(P<0.05).Conclusion:Under hypoxic conditions,the expression of Aβ1-42 and BACE-1 protein in PC12 cells increased.Sesamin reduced the expression of Aβ1-42 and BACE-1 that increased under hypoxia.
