摘 要:目的:观察三黄固本汤对激素干预下系统性红斑狼疮(SLE)小鼠骨密度、骨矿含量及脂代谢的影响,进一步研究探索其防治激素性股骨头坏死的实验室微观证据。方法:选择8~10周的雌性MRL/lpr小鼠30只,按照随机数字表分为三组,即模型组、对照组(泼尼松组)、治疗组(泼尼松+中药组),每组各10只。其中治疗组和对照组先以泼尼松饲养7 d,待其进入SLE慢性缓解期后,治疗组再加入中药进行干预,8周后用小动物双能X线骨密度仪对各组小鼠治疗后骨密度及骨矿含量进行测定。同时针对脂肪代谢相关指标对各组小鼠治疗前后进行测定比较。结果:基于骨密度、骨矿含量、TG、CH、HCL的测定,对照组与模型组比较,差异具有统计学意义(均P<0.05);治疗组治疗后与模型组比较,差异无统计学意义(P>0.05);对照组与治疗组治疗后比较,差异有统计学意义(P<0.05)。结论:三黄固本汤可以防止使用激素干预治疗下的SLE小鼠骨密度及骨矿含量出现减少,并且可调节脂代谢相关指标,其防治股骨头坏死的作用机制有可能是通过此靶点而实现的。Objective:To observe the effects of Sanhuang Guben decoction on bone density,bone mineral content and lipid metabolism in mice with systemic lupus erythematosus(SLE)under hormone intervention.Methods:Thirty female MRL/lpr mice were selected for 8~10 weeks and were divided into 3 groups according to the random number table,namely,model group,control group(prednisone group),treatment group(prednisone traditional Chinese medicine group),each group was 10.The treatment group and the control group were raised with prednisone for 7 days.After entering the SLE chronic remission stage,the treatment group then added traditional Chinese medicine to intervene for 8 weeks.The bone density and bone mineral content of mice in each group were determined by small animal dual energy X line bone density instrument.At the same time,the related indexes of fat metabolism were compared before and after treatment.Results:On the basis of the determination of bone mineral density,bone mineral content and TG、CH、HCL,the control group and the model group had obvious changes,the difference was statistically significant(P<0.05).The treatment group and the model group had no significant changes the difference wasn’t statistically significant(P>0.05).The control group and treatment group had obvious changes,the difference was statistically significant(P<0.05).Conclusion:This study shows that Sanhuang Guben decoction can prevent the decrease of bone mineral density and bone mineral content in SLE mice treated with hormone intervention,and can regulate the related indexes of lipid metabolism.The mechanism of prevention and treatment of femoral head necrosis may be achieved by this target.
