摘 要:为了深入探究钙依赖蛋白激酶(Calcium dependent protein kinase,CDPK)在小麦与叶锈菌互作过程中的功能机制,本试验结合叶锈菌生理小种260侵染小麦品种Tc Lr26后的RNA-seq数据和NCBI基因组数据筛选得到抗病相关基因TaCDPK6,并对TaCDPK6进行了蛋白质理化性质分析;利用RT-qPCR和Western blotting技术检测TaCDPK6基因及其表达蛋白在小麦与叶锈菌互作过程中的表达模式。结果表明,TaCDPK6蛋白分子大小为57 kD,理论等电点为7.61,为亲水性蛋白质;在小麦叶片接种叶锈菌后,随着接种时间的延长,目的基因及蛋白呈先上升后下降的表达趋势,并且在4~8 h表达量达到最高,与RNA-seq数据分析得到的表达趋势基本一致。结果表明TaCDPK6在小麦抵抗叶锈菌的基础防御反应中可能发挥正调控作用。The research aims to further explore the functional mechanism of Calcium dependent protein kinase(CDPK) in the interaction between wheat and Puccinia triticina. In this study, the resistance gene TaCDPK6 was screened by combining the RNA-seq data and NCBI genomic data of Tc Lr26 infected by the physiological race 260 of Puccinia triticina, and the physicochemical properties of TaCDPK6 protein were analyzed. RT-qPCR and Western Blotting were used to detect the expression pattern of TaCDPK6 gene and its expressed protein during the interaction between wheat and Puccinia triticina. The results showed that the molecular size of TaCDPK6 was 57 kD and the theoretical isoelectric point was 7.61, which indicated that TaCDPK6 was a hydrophilic protein. With the extension of inoculation time, the expression trend of target gene and protein was firstly increased and then decreased after inoculation, and the expression level reached the highest at 4-8 h, which was basically consistent with the expression trend obtained from RNA-seq data analysis. The results show that TaCDPK6 plays a positive role in the basic defense response of wheat against Puccinia triticina.
