摘 要:为了解传染性支气管炎病毒S1基因遗传变异情况,用鸡胚传代、血凝特性检测、传染性支气管炎病毒(Infectiousbronchitisvirus,IBV)膜蛋白(Membrane,M)基因检测等方法对保定地区鸡群分离的9株病毒进行了鉴定,并对其S1蛋白基因进行了遗传变异分析。结果表明:9株分离毒接种鸡胚尿囊液直接血凝均为阴性,间接血凝和IBV M基因检测均呈阳性。9株IBV分离毒S1蛋白基因编码区长1608/1611/1620 nt,分别编码536/537/540个氨基酸;与27株IBV参考毒株间的核苷酸和氨基酸序列同源性分别为59.8%~99.3%和49.2%~98.9%。9株分离毒S蛋白裂解位点模式为HRRRR/HRRKR/HRHRR。9株分离毒均属于Ⅰ群中的LX4型IBV,与疫苗毒YX10D90vaccine株亲缘关系较近,与疫苗毒LDT3-A、4/91vaccine、Ark99、J9、Connecticut vaccine、H120株等亲缘关系较远,与疫苗毒Georgia 1998 Vaccine株亲缘关系最远。本研究结果可为传染性支气管炎防控提供有益参考。To investigate the genetic variation of infectious bronchitis virus S1 gene,9 isolates isolated from Baoding area were identified by methods such as chicken embryo passage,hemagglutination characteristics detection,and IBV membrane protein gene detection.The genetic variation of IBV S1 protein gene was analyzed.The results showed that the chicken embryo allantoic fluid of the 9 isolates was negative for direct hemagglutination assay,while was positive both for indirect hemagglutination assay and IBV M gene detection.The full length of coding region of the S1 protein gene of the isolates comprised of 1608/1611/1620 nucleotide,which encoding 536/537/540 amino acids,respectively.The result of sequence alignment of S1 gene showed that the nucleotide and amino acid homology between 9 isolates and 27 reference strains were 59.8%-99.3%and 49.2%-98.9%,respectively.The S protein cleavage recognition sites of the isolates were HRRRR,HRRKR and HRHRR.The results of phylogenetic analysis of 9 isolates and 27 reference strains showed the isolates all belonged to IBV of LX4 type in group I.The isolates were closely related to vaccine strain YX10 D90,and had relatively far evolutionary relationship with vaccine strain e.g.LDT3-A,4/91 vaccine,Ark99,J9,Connecticut vaccine,and H120.The genetic relationship between the isolates and Georgia 1998 Vaccine strain was the most distant.The results of this study can provide a useful reference for the prevention and control of infectious bronchitis.
