摘 要:目的生姜提取物6-姜酚在创面修复中的作用研究较少。文章拟探讨在创面愈合过程中生姜提取物6-姜酚对相关因子如转化生长因子β1(TGF-β1)、血小板衍生生长因子(PDGF)、血管内皮生长因子(VEGF)以及增殖细胞核抗原(PCNA),α-平滑肌肌动蛋白(α-SMΑ)的影响,并尝试探讨其机制。方法在C57BL/6小鼠的脊柱两侧对称性地各做一个直径8 mm的圆形创面模型,设置正常组(只注射等渗盐水)、6-姜酚治疗组[局部注射6-姜酚,30 mg/(kg·d)],伤口周围局部注射药物进行干预治疗。采用HE染色检测创面组织病理变化;分别采用免疫组织化学法和免疫荧光法检测创面中PCNA、α-SMA和MPO、F4/80的表达变化;采用蛋白质免疫印迹技术和实时荧光定量技术检测创伤组织内TGF-β1、PDGF、VEGF以及PCNA、α-SMA的蛋白质和mRNA的表达水平。结果术后第5天,与正常组上皮间隙[(1.68±1.17)mm]相比,HE染色显示6-姜酚治疗组的上皮间隙[(0.77±0.43)mm]降低(P<0.05);与正常组PCNA、α-SMΑ、F4/80的阳性细胞数相比,6-姜酚治疗组的阳性细胞数均升高(P<0.05)。Western blot和RT-qPCR检测结果显示,与正常组PDGF、VEGF、TGF-β1、PCNA、α-SMΑ表达量相比,6-姜酚治疗组均升高(P<0.05)。术后第5天,6-姜酚治疗组小鼠羟脯氨酸含量(3.51±1.04)较正常组(2.31±0.84)显著提高(P<0.01)。结论6-姜酚可通过调节部分局部生长因子和PCNA、α-SMA的表达进而促进小鼠创面愈合。Objective The effect of ginger 6-gingerol in wound healing is seldom studied.This article aims to explore the effect and mechanism of ginger 6-gingerol on related factors such as TGF-β1,PDGF,VEGF,PCNA andα-SMA in the process of wound healing.Methods A 8 mm circular wound was symmetrically made on both sides of the spine of a C57BL/6 mouse,and the normal model(MOD)group was set.The 6-gingerol(GIN,30 mg/kg/d)drugs was locally injected around the wound in the treatment group.HE staining was used to detect the pathological changes in wound tissue;Immunohistochemistry and immunofluorescence were respectively used to detect the expression changes of PCNA,α-SMA,MPO and F4/80 in wounds;Western blotting and real-time fluorescence quantitative techniques were used to detect PDGF,VEGF,TGF-β1,PCNA,α-SMΑin wounds.Results On the 5th day after operation,HE staining showed that the epithelial space[(0.77±0.43)mm]of the 6-gingerol treatment groups were reduced compared with the epithelial space[(1.68±1.17)mm]of the normal model groups(P<0.05);Compared with the number of the positive cells of PCNA,α-SMA,and F4/80 in the normal model groups,the number of the positive cells in the 6-gingerol treatment group increased(P<0.05).Western blot results showed that the expression of PDGF,VEGF,TGF-β1,PCNA,andα-SMA in the normal model groups were all increased in the 6-gingerol treatment group(P<0.05).RT-qPCR detection results showed that the expression of PDGF,VEGF,TGF-β1,PCNA,andα-SMA in the normal model group was all increased in the 6-gingerol treatment group(P<0.05).Conclusion The 6-gingerol can promote wound healing in mice by regulating the expression of some local growth factors,PCNA andα-SMA.
