摘　　要：利用不同浓度的Erastin处理人肺癌A549和A549/DDP细胞,以探究其对MACC1基因表达的影响.采用qRT-PCR,Western Blot检测MACC1 mRNA和蛋白在A549,A549/DDP细胞中表达的差异性,以及不同浓度的Erastin对MACC1基因表达的影响.结果表明,不同浓度的Erastin处理人肺癌A549和A549/DDP细胞48 h后,2种细胞的生长均被明显抑制,与对照组相比终浓度为5,40μmol/L的Erastin处理组其MACC1 mRNA和蛋白表达水平显著降低.在0~40 mg/L终浓度范围内,Erastin能显著降低A549/DDP细胞内MACC1 mRNA和蛋白的表达.Human lung cancer A549 and A549/DDP cells were treated with different concentrations of erastin to explore their effects on MACC1 gene expression.The expression of MACC1 mRNA and protein in A549 and A549/DDP cells and the effect of different concentrations of Erastin on MACC1 gene expression were detected by QRT-PCR and Western Blot.The resultsthow that human lung cancer A549 and A549/DDP cells were treated with different concentrations of Erastin for 48 h,and the growth of both cells was significantly inhibited.The expression levels of MACC1 mRNA and protein were significantly decreased in the 5,40μmol/L Erastin treatment groups.Erastin can significantly decrease the expression of MACC1 mRNA and protein in A549/DDP cells in the final concentration range of 0~40 mg/L.
