摘  要:烟酰胺腺嘌呤二核苷酸磷酸-苹果酸酶(NADP-malic enzyme,NADP-ME)由小基因家族编码,在植物生长发育和应对逆境胁迫中发挥重要作用。为了进一步筛选甜菜NADP-ME基因并分析其功能,本研究对该家族成员进行了详细的生物信息学分析。结果显示,甜菜基因组共有5个NADPME基因,基因长度介于6537 bp~19759 bp,并且所有BvNADP-ME均为亲水性蛋白,主要定位于线粒体、质膜、内质网和细胞核上,无信号肽,不存在跨膜结构域,不属于跨膜蛋白。除BvNADP-ME5外,其它家族成员均为酸性蛋白质;其中BvNADP-ME2、3、4皆为不稳定蛋白;导肽分析表明BvNADP-ME3含有叶绿体转运肽,BvNADP-ME4、5均含有线粒体靶向肽。染色体定位分析发现,BvNADP-ME5、2、1主要分布在1、2、8号染色体上。蛋白二三级结构预测具有相似的特征,α-螺旋和无规则卷曲为BvNADP-MEs的主要结构元件。系统进化分析表明BvNADP-MEs可分为6个亚组(I~VI),其中甜菜与菠菜NADP-ME亲缘关系最近,其次是藜麦,最远是拟南芥。蛋白质保守基序分析表明甜菜NADP-ME家族蛋白包含14个基序,其中Motif4、Motif6、7、8和Motif11、12可能与烟酰胺腺嘌呤二核苷酸(NAD)的结合有关。分析BvNADP-ME基因家族启动子区域的顺式作用元件,结果显示各成员启动子均含有植物激素响应元件和非生物胁迫响应元件。本研究对5个BvNADP-ME基因家族成员的生物信息学进行分析,为深入研究甜菜NADP-ME基因功能提供参考依据,也为分子抗性育种以及提高甜菜的品质和产量提供新思路。NADP-malic enzyme(NADP-ME),encoded by a small gene family,plays an important role in plant growth and development and responses to stress.In order to screen the BvNADP-ME genes and analyze their functions,we carried out a detailed bioinformatics analysis on the members of BvNADP-MEs family.The results showed that there were five NADP-ME genes in beet genome,the gene length was 6537 bp-19759 bp,and all BvNADP-ME were hydrophilic proteins,mainly located in mitochondria,plasma membrane,endoplasmic reticulum and nucleus,without signal peptide,transmembrane domain and transmembrane protein.Except for BvNADP-ME5,other members of the family were acidic proteins;among them,BvNADP-ME2,3 and 4 were all unstable proteins;the results showed that BvNADP-ME3,4 and 5 contained chloroplast transport peptide,mitochondrial targeting peptide and mitochondrial targeting peptide,respectively.Chromosomal localization analysis found that BvNADP-ME 5,2 and 1 were mainly distributed on chromosomes 1,2 and 8.The prediction of protein secondary and tertiary structure had similar characteristics,α-Helix and random coil were the main structural elements of BvNADP-MEs.Phylogenetic analysis showed that BvNADP-MEs could be divided into six subgroups(I~VI).Among them,beet and spinach had the closest genetic relationship with NADP-ME,followed by quinoa and Arabidopsis.The analysis of protein conserved motifs showed that there were 14 motifs in beet NADP-ME family proteins.Motif 4,Motif 6,Motif 7,Motif 8 and Motif 11,Motif 12 may be related to NAD binding.The cis acting elements in the promoter region of BvNADPME gene family were analyzed.The results showed that each promoter contained plant hormone response elements and abiotic stress response elements.The analysis of the bioinformatics of five NADP-ME genes provided a reference for further study on the function of NADP-ME gene in sugar beet,and also provided a new idea for molecular breeding for resistance and quality and yield of sugar beet.
