摘 要:为了筛选适用于甜菜品种鉴定的DAMD分子标记,建立高效的甜菜品种鉴定方法,利用从文献中获得的29条DAMD引物对40个甜菜品种进行PCR扩增。结果表明:URP1F、OGRB01、URP4R、URP2R、URP6R、URP13R、URP32F、URP17R具有高多态性,8条引物总计可以扩增101条带,其中99条为多态性条带,多态性百分比为97.22%,可确定为鉴定甜菜品种的核心引物。根据扩增产物,可采用特征谱带法,单独使用引物URP6R或引物URP1F和引物URP17R共同使用鉴别40个甜菜品种。同时根据扩增产物建立0、1数据库计算出品种间遗传距离范围为0.01~0.25,在遗传距离为0.25时,可将40个甜菜品种分为2个类群。类群之下可分成多个亚群,通过遗传距离建立的聚类图也可将甜菜品种逐一鉴别。研究表明基于DAMD引物建立的高多态性指纹图谱对今后鉴别甜菜品种的真实性提供了高效快捷的方法。In order to screen DAMD molecular markers suitable for beet variety identification,an efficient sugar beet variety identification method was established.In this study,29 DAMD primers obtained from the literature were used for PCR amplification of 40 sugar beet varieties.The results showed that URP1F,OGRB01,URP4R,URP2R,URP6R,URP13R,URP32F and URP17R had high polymorphism,and a total of 101 bands could be amplified with 8 primers,of which 99 were polymorphic bands with a polymorphism percentage of 97.22%,which could be identified as the core primer for identifying beet varieties.According to the amplification products,40 beet varieties could be identified by using primer URP6R alone or primer URP1F and primer URP17R together by using characteristic spectral band method.At the same time,the range of genetic distance between varieties was calculated as 0.01-0.25 according to the establishment of 0 and 1 database of amplification products.When the genetic distance was 0.25,40 varieties could be divided into 2 groups.The groups could be divided into several subgroups,and the beet varieties could be identified one by one by the cluster map established by genetic distance.The study showed that the high polymorphism fingerprint based on DAMD primers will provide an efficient method for identifying the authenticity of sugar beet varieties in the future.
