摘 要:Na^(+)/H^(+)逆向转运蛋白(NHX)在植物响应盐胁迫和生长发育过程中发挥着重要的调控作用。本研究对菠菜NHX家族基因进行全基因组鉴定和生物信息学分析,并利用qRT-PCR技术检测NHX基因在盐胁迫下的表达。结果显示,从菠菜基因组中共鉴定出6个SpoNHXs基因,系统进化树分析发现其分属3个亚家族——Vac、Endo和PM;这6个SpoNHXs均具有跨膜结构域,与拟南芥NHX家族成员在不同亚家族中的保守基序高度一致,表明其具有相似的生物学功能;这6个SpoNHXs受盐胁迫上调表达,其中SpoNHX1与SpoNHX6显著上调。本研究结果可为后续SpoNHXs基因克隆和耐盐功能分析提供理论支持。Na^(+)/H^(+) antiporter(NHX) family genes play important regulatory roles in plant response to salt stress and growth and development. Thus, in this study, the genome-wide identification of NHX genes in spinach and bioinformatics analysis were carried out, and the expression patterns under salt stress were detected by qRT-PCR. The results showed that six SpoNHXs genes were identified in spinach genome. Phylogenetic analysis showed that they belonged to three subfamilies(Vac, Endo and PM). The six SpoNHXs had transmembrane domains, which were highly consistent with the conserved motifs of Arabidopsis NHX family members in different subfamilies, indicating that they had similar biological functions. The expressions of the six SpoNHXs were up-regulated under salt stress, among which, SpoNHXs and SpoNHX6 were significantly up-regulated. The results of this study could provide references for further cloning of SpoNHXs genes and analysis of salt tolerance function.
