摘 要:为了探究辐射对柱花草(Stylosanthes spp.)M_(3)代农艺性状的影响及分子标记辅助诱变育种技术,利用经^(60)Coγ射线辐射诱变后的柱花草种子,种植后经过连续2代的单株选择筛选到17份性状优良的M_(2)代柱花草种子,种植成为M3代后,对17份柱花草测定其株高、冠径、分枝数、第一分支长、病害级数、开花期、单株干重,并进行DNA保守序列多态性(Conserved DNA-derived polymorphism,CDDP)和相关序列扩增多态性(Sequence related amplified polymorphism,SRAP)的遗传多样性分析。运用NTSYS-pc 2.1软件计算17份柱花草的遗传相似系数,17条CDDP引物以及26对SRAP引物分别扩增出92条和177条带,其中多态性条带数分别为66条和111条,多态性比率分别为71.7%和62.7%,特异性条带各为13条和29条,特异性比率分别为14.1%和16.4%,种质间的遗传相似系数(GS)范围为0.54~0.90和0.60~0.94,平均遗传相似系数为0.76和0.79。按照非加权配对算术平均法基于CDDP标记和SRAP标记联合作17份柱花草的聚类图,在遗传相似系数为0.82时可将17份柱花草分成7个类群,聚类结果与农艺性状统计结果基本一致。本研究为今后辐射诱变柱花草育种提供一定的参考依据。To explore the effects of radiation on the agronomic traits of Stylosanthes spp.M_(3) generation and molecular marker-assisted mutation breeding technology,plant height,crown diameter,number of branches,first branch length,disease grade,flowering stage and dry weight per plant were measured and genetic diversity analysis of conserved DNA-derived polymorphism and sequence related amplified polymorphism was made by using 17 samples of M3 generation Stylosanthes spp.seeds that were mutagenized by ^(60)Coγ-ray radiation and selected with excellent traits after 2 consecutive generations of single plant selection from M_(2) generations.The genetic similarity coefficients of 17 Stylosanthes spp.were calculated using NTSYS-pc 2.1 software,as a result,92 and 177 bands were amplified by 17 CDDP primers and 26 pairs of SRAP primers respectively,in which the number of polymorphic bands were 66 and 111 respectively,the polymorphism rates were 71.7%and 62.7%,the specific bands were 13 and 29,and the specificity rates were 14.1%and 16.4%respectively.The genetic similarity coefficient(GS)between germplasms ranged from 0.54~0.90 and 0.60~0.94,and the average genetic similarity coefficient were 0.76 and 0.79 respectively.The cluster map of 17 Stylosanthes spp.showed the 17 Stylosanthes spp.could be divided into 7 groups and the clustering results were basically consistent with the statistical results of agronomic traits when the genetic similarity coefficient was 0.82 according to the unweighted paired arithmetic average method and made based on the combination of CDDP and SRAP markers.This study provides a certain reference basis for the breeding of Stylosanthes spp.by radiation mutation in the future.
