摘 要:为了研究长途运输对猪卵母细胞体外成熟及后续发育的影响,试验对长途运输及长途运输后不同培养方式培养的猪卵母细胞成熟率、核移植后重构胚和孤雌激活胚的囊胚率进行了测定,并以重庆市畜牧科学院生物工程研究所往期数据为对照进行比较分析。结果表明:长途运输后的猪卵母细胞成熟率为81.20%,与对照组(82.14%)比较差异不显著(P>0.05);长途运输后采用圆底玻璃管直接培养的猪卵母细胞成熟率为87.00%,显著高于用4孔细胞培养板培养的猪卵母细胞成熟率(77.33%,P<0.05)。经长途运输后猪卵母细胞通过核移植后重构胚的囊胚率为19.20%,与对照组(20.28%)比较差异不显著(P>0.05);经长途运输后成熟卵母细胞孤雌激活胚的囊胚率为23.65%,极显著低于对照组的38.05%(P<0.01)。说明长途运输不影响猪卵母细胞的成熟,但是会影响其后续发育。To study the effect of long-distance transport on in vitro maturation and subsequent development of porcine oocytes, the maturation rate of oocytes and the blastocyst rate of subsequent parthenogenetic activated embryos and reconstructed embryos after nuclear transfer were measured in pigs cultured by long-distance transportation and different culture methods, and the data of bioengineering institute of Chongqing Academy of Animal Science in the past were used as control. The results showed that the maturation rate of porcine oocytes after long-distance transportation was 81.20%, which was not significantly different from that of the control group(82.14%,P>0.05). After long-distance transportation, the maturation rate of porcine oocytes directly cultured in round-bottomed glass tubes was 87.00%, which was significantly higher than that of porcine oocytes cultured in four-hole cell culture plates(77.33%, P<0.05). After long-distance transport, the blastocyst rate of reconstructed embryos in porcine oocytes after nuclear transfer was 19.20%, which was not significantly different from that of control group(20.28%, P>0.05), and the blastocyst rate of parthenogenetic activated embryos in porcine oocytes was 23.65%, which was extremely significantly lower that of the control group(38.05%, P<0.01). The results indicated that long-distance transport did not affect the maturation of porcine oocytes, but it affected its subsequent development.
