摘  要:为了研究梅花鹿接种卡介苗前后信号通路的变化情况,取接种卡介苗(DF组)和未接种卡介苗(SY组)的梅花鹿外周血进行转录组测序,构建mRNA文库,利用DESeq软件对获得的mRNA进行差异表达分析,根据分析结果,选取免疫相关表达上调基因进行qPCR验证。结果表明:DF组和SY组共有1 337个显著差异表达的基因,其中上调基因658个,下调基因679个;差异基因主要参与机体应激、信号转导、调节白细胞迁移等生物学过程。梅花鹿接种卡介苗后主要富集到PI3K-Akt、Notch、NF-κB、Toll样受体以及细胞外基质受体相互作用等免疫相关的信号通路。qPCR检测结果与高通量测序结果一致。说明接种卡介苗后,梅花鹿体内的蛋白质及细胞外基质修饰过程得到了增强,受体活性肽、转录因子的结合能力和细胞因子活性增强。In order to study the changes of signal pathway before and after BCG vaccine inoculation in sika deer, the transcriptome sequencing was performed on the peripheral blood of sika deer vaccinated with BCG vaccine(DF group) and unvaccinated with BCG vaccine(SY group), and mRNA library was constructed. The mRNA differential expression analysis was performed on the obtained mRNAs by using the DESeq software. According to the analysis results, the immune-related up-regulated genes were selected for qPCR verification. The results showed that there were 1 337 significantly differentially expressed genes in DF group and SY group, among which 658 genes were up-regulated and 679 genes were down-regulated. Differential genes were mainly involved in biological processes such as stress, signal transduction and regulation of leukocyte migration. After BCG vaccine inoculation, sika deer were mainly enriched in immune-related signaling pathways such as PI3 K-Akt, Notch, NF-κB, Toll-like receptor and extracellular matrix receptor interaction. The results of qPCR were consistent with those of high-throughput sequencing. These results indicated that after inoculation with BCG vaccine, the modification process of protein and extracellular matrix in sika deer was enhanced, and the binding ability of receptor active peptide, transcription factor and cytokine activity were enhanced.
