摘  要:为阐明高原牦牛适应低氧环境时肺脏血管低氧相关因子在不同发育阶段表达变化的特点,试验选取1日龄、30日龄、180日龄和成年四个发育阶段的大通牦牛,运用免疫组织化学方法,对低氧诱导因子1α(Hif-1α)、血管内皮生长因子(VEGF)和诱导型一氧化氮合酶(iNOS)在肺脏组织中的表达进行测定。结果表明:随着年龄增长,大通牦牛肺脏组织中Hif-1α的阳性率表现为先升高后降低再升高的特点,各发育阶段之间差异显著(P<0.05);Hif-1α的平均光密度值表现为先升高后降低再升高的趋势,除1日龄和180日龄差异不显著(P>0.05)外,其余各发育阶段之间差异显著(P<0.05)。肺脏组织中VEGF的阳性率和平均光密度值均表现为先降低再升高的趋势,各发育阶段之间差异显著(P<0.05)。iNOS的阳性率表现为先降低再升高的趋势,各发育阶段之间差异显著(P<0.05);平均光密度值随着年龄增长,表现为先降低再逐渐升高的趋势,除180日龄与其他各发育阶段均差异显著外(P<0.05),其余各发育阶段之间差异不显著(P>0.05)。说明Hif-1α对下游基因的调控和下游基因的转录、翻译可能不同步。To clarify the expression changes of pulmonary vascular hypoxia-related factors at different developmental stages of plateau yaks when they adapt to hypoxic environment, four developmental stages of 1-day-old, 30-day-old, 180-day-old and adulthood Datong yaks were selected as the research objects. The expressions of the hypoxia-inducible factor-1α(Hif-1α), vascular endothelial growth factor(VEGF), and inducible nitric oxide synthase(iNOS) in lung tissue were measured by immunohistochemical method. The results showed that the positive rate of the Hif-1α in the lung tissue of the yaks increase at first, then decrease, and increase again with age, and the difference was significant among different developmental stages(P<0.05). The average optical density of Hif-1α showed a trend of increasing at first, then decreasing and then increasing, and there were significant differences among developmental stages(P<0.05) except that there was no significant difference at 1 day of age and 180 days of age(P>0.05). The positive rate and average optical density of VEGF in lung tissue showed a trend of decreasing first and then increasing, with significant differences among different developmental stages(P<0.05). The positive rate of iNOS showed a trend of decreasing first and then increasing, and there was significant difference among different developmental stages(P<0.05). The average optical density first decreased and then gradually increased with age, there was significant difference between 180 days of age and other developmental stages(P<0.05), but no significant difference between other developmental stages(P>0.05). It indicated that Hif-1α regulation of downstream genes might be out of sync with transcription and translation of downstream genes.
