摘  要:目的:观察不同高原低氧暴露时间下大鼠肺泡组织细胞自噬变化情况,探讨其在高原低氧性肺损伤中的作用。方法:将雄性Wistar大鼠40只随机分为平原对照组(CG,陕西省西安市,海拔400m)及高原低氧1天、7天、14天、21天组(HG,青海省玉树州杂多县,海拔4300m),共5组,大鼠麻醉后取右肺组织,HE染色观察肺组织大体形态变化,电镜观察肺泡组织细胞超微结构及自噬情况,Western-blot法检测肺组织自噬特异性蛋白Beclin-1及LC3-Ⅱ的表达水平。结果:HE染色显示,与平原对照组比较,高原低氧组随着暴露时间的延长,大鼠肺泡组织细胞损伤程度逐渐加重;电镜结果显示,随着低氧暴露时间的延长,两组肺泡组织细胞超微结构损伤破坏逐渐加重,并出现自噬样结构,自噬程度逐渐增加;与平原对照组比较,高原低氧各组大鼠肺泡组织Beclin-1及LC3-Ⅱ的蛋白表达水平均随着低氧暴露时间的延长而逐渐升高(P<0.05)。结论:进入高原低氧环境后,大鼠肺泡组织细胞自噬水平随低氧暴露时间的延长而逐渐升高,同时肺泡组织细胞损伤程度也逐渐加重,自噬水平升高可能与高原低氧性肺损伤密切相关。Objective:The objective of this study is to examine the autophagy of alveolar cells exposed to high altitude hypoxia for different time periods and its roles in lung injury.Methods:40 male Wistar rats were randomized into 5 groups,which were exposed to an altitude of 400 m in Xi’an(CG),or to a high altitude of 4300 m in Qinghai Yushu Zaduo for 1 days,7 days,14 days and 21 days.Lung tissues were obtained and stained with hematoxylineosin for observations of the ultrastructure of alveolar cells and autophagy by transmission electron microscopy,and Beclin-1 and LC3-Ⅱprotein levels in alveolar tissues were determined by Western-blot respectively.Results:Compared to control group,exposure to high altitude hypoxia for longer time periods results in aggravation of lung injury,formation of more autophagosomes,and alveolar cell injury became more severe as the exposure duration increased.Compared to control rats,the protein levels of Beclin-1 and LC3-Ⅱin rats exposed to hypoxia for 1 days,7 days,14 days and 21 days were significantly increased,and the longer the exposure duration will be,the greater the protein expression increases(P<0.05).Conclusion:Exposure to high altitude hypoxia for longer time periods can induce autophagy of alveolar cells and can result in aggravation of lung injury,which may be an important mechanism of high altitude hypoxia induced lung injury.
