摘 要:高丹草(Sorghum bicolor×S.sudanense)是一种优质牧草。为获得高丹草多倍体材料,试验以秋水仙素为诱导剂,以引进的高丹草品种‘大卡’为供试材料,应用浸种法和滴生长点法分别对高丹草种子和幼苗生长点进行了处理,对获得的诱变后代采用表型测定、根尖染色体法和表皮气孔保卫细胞法相结合的方式,通过逐代鉴定筛选,最终在M5代获得了稳定遗传的两份高丹草多倍体材料。本研究可为高丹草多倍体诱导相关研究提供参考;同时创制的两份多倍体材料除可作为种质资源使用外,经进一步培育后有望作为品种在生产上推广应用。Sorghum bicolor×S.sudanense is a kind of elite forage grass.To obtain Sorghum bicolor×S.sudanense polyploids,colchicine was used as chemical mutagen,‘Big Kahuna’as material,the seeds and seedling growth points were treated by the method ofseed-immersion method and drop growth points,respectively.The mutation polyploid progenies of Sorghum bicolor×S.sudanensewere identified by combining the methods of phenotyping,root tip chromosome and epidermal stomatal guard cells.After identification and screening generation by generation,two polyploids obtained in M5 generationweregenetically stable.This study would provide reference for the relevant research of Sorghum bicolor×S.sudanense polyploid induction;and two polyploids created in the study not only can be applied as germplasms,but also are expected to be varieties by further cultivation for promotion and application in grass production.
