摘 要:采用同时蒸馏萃取法(SDE)提取两种不同产地的奇异南星挥发油,通过气相色谱-质谱联用仪鉴定其中化学成分,采用峰面积归一化法确定各组分的相对含量,并采用甲基噻唑蓝(MTT)法对两种挥发油进行体外抗肿瘤活性测试。结果显示,从广西产和四川产奇异南星挥发油中分别鉴定出12种和11种化合物,其相对含量分别为90.96%和94.68%,相同成分有5种,分别为亚油酸、棕榈酸、十五碳酸、5-十八碳炔和顺-11-十六碳烯酸;两种挥发油主要成分为脂肪酸,但组成和含量存在差异。两种挥发油对人肺癌细胞株A549、人宫颈癌细胞株Hela、人肝癌细胞株Hep G2和人结肠癌细胞株HCT-116均显示出较强的抑制活性,其细胞毒性无明显差异,半抑制浓度范围为43.66~105.04 mg/L。Two kinds of volatile oils of Arisaema decipiens Schott from two habitats were extracted by simultaneous distillation-extraction(SDE), and their chemical components were identified by gas chromatography-mass spectometry(GC-MS). The relative contents of each component were determined by peak area normalized method. The antitumor activity of the two volatile oils in vitro was tested by 3-(4,5)-dimethylthiahiazo(-z-y1)-3,5-di-phenytetrazoliumromide(MTT) assay. The results showed that twelve and eleven kinds of volatile components from Arisaema decipiens Schott from Guangxi and Sichuan were identified, respectively. And their relative contents calculated by GC peak areas were 90.96% and 94.68%, respectively. There were five kinds of the same components, namely(Z,Z)-9,12-octadecadienoic acid, n-hexadecanoic acid, pentadecanoic acid, 5-octadecyne and Z-11-hexadecenoic acid. The main components of two volatile oils from Arisaema decipiens Schott were fatty acids, but they were different in the component and relative contents. Both volatile oils inhibited cellular proliferation significantly against the human lung cancer cell line A549, human cervical cancer cell line Hela, human liver cancer cell line HepG2 and human colon cancer cell line HCT-116. There was no significant difference in cytotoxicity, and half maximal inhibitory concentration ranged from 43.66 to 105.04 mg/L.
