瑶药多穗柯提取物对糖尿病大鼠基质金属蛋白酶8、9的影响作用研究

摘  要:目的探讨瑶药多穗柯的降糖作用及作用机制。方法采用高脂饲料+链脲菌素(STZ)制备大鼠糖尿病模型,通过瑶药多穗柯提取物灌胃进行药物干预,检测大鼠体重、血糖、血清胰岛素(INS)、血清糖化血红蛋白(GHbA1c)、血清基质金属蛋白酶8(MMP-8)及基质金属蛋白酶9(MMP-9)水平和胰腺病理等指标,从而观察多穗柯降糖作用。结果与模型组比较,多穗柯提取物7 g生药/kg剂量组大鼠体重增加,血糖降低,血清糖化血红蛋白及基质金属蛋白酶9水平降低,血清胰岛素水平升高,胰腺病理病变明显改善。结论瑶药多穗柯7 g生药/kg剂量具有显著降糖作用,其机制可能与改善胰岛功能,促进胰岛素分泌,调控糖化血红蛋白与基质金属蛋白酶9水平相关。Objective To study the hypoglycemic effect of Lithocarpus polystachyus and its mechanism.Methods Diabetic rat model induced by high-fat diet feeding and Streptozotocin Injection,Through giving the Lithocarpus polystachyus extract to observe the effect of polycotyledon extract on body weight,blood glucose,serum insulin,serum glycated hemoglobin,serum matrix metalloproteinase 8,serum matrix metalloproteinase 9 and pathological changes of pancreas in rats.Results Compared with the model group,Lithocarpus polystachyus extract which dosage was 7 g crude drug/kg could increase the weight of diabetic rats,decrease the levels of fasting blood glucose,serum GHbA1c and serum MMP-9,increase the levels of serum INS,and improve the changes of pancreatic tissue.Conclusion Lithocarpus polystachyus has significant hypoglycemic effect,which may be related to the improvement of islet function,insulin secretion,regulation of glycated hemoglobin and MMP-9 level.
