摘 要:目的:探讨癌痛消方对肝癌大鼠癌痛的调控作用机制。方法:采用肝脏左叶表面注射walker256细胞成功构造60只原发性肝癌SD大鼠模型,根据癌痛消方剂量分为0.5 g/mL组、1.0 g/mL组、2.0 g/mL组,每组20只,并设置对照组(Control组,n=20)。分别于给药第5、10、15天,采用Weternblot检测各组大鼠的核因子kB(NF-kB)和单一免疫球蛋白白细胞介素1受体相关蛋白(SIGIRR)水平;采用逆转录聚合酶链反应(RT-PCR)检测脊髓小胶质细胞标志性CD11b和Toll受体2(TLR2)及受体4(TLR4)mRNA的表达水平;采用流式细胞仪检测细胞周期和凋亡情况。结果:Western blot实验结果显示,癌痛消方的浓度越大,NF-kB蛋白的表达水平越低,SIGIRR蛋白的表达水平越高(均P<0.05)。RT-PCR实验结果显示,癌痛消方的浓度越大,TLR2、TLR4 m RNA的表达水平越低(均P<0.05)。流式细胞仪检测结果显示,癌痛消方的浓度越大,G0/G1期细胞比例越高,S期、G2期细胞比例越低(均P<0.05);细胞凋亡率也越高(均P<0.05)。结论:癌痛消方能够减轻肝癌小鼠疼痛程度,促使肝癌细胞凋亡,其机制可能与调控调控TLR-NF-kB通道有关。Objective: To explore the regulatory effect and mechanism of Aitongxiao prescription on cancer pain in rats with liver cancer. Methods: Sixty primary liver cancer SD rats models were established by injecting Walker 256 cells into the left lobe of liver. According to the dosage of Aitongxiao prescription, the rats were divided into 0.5 g/mL group, 1.0 g/mL group and 2.0 g/mL group, with 20 rats in each group, and 20 normal rats were defined as a control group. The protein expression levels of nuclear factor κB(NF-κB) and single immunoglobulin interleukin-1 receptor associated protein(SIGIRR) were detected by Western blot on the 5 th, 10 th and 15 th day after administration. RT-qPCR was used to detect the mRNA expressions of Toll receptor(TLR) 2 and TLR4.Flow cytometry was used to detect cell cycle and apoptosis. Results: The higher the dose of Aitongxiao prescription, the lower the expression of NF-κB protein, the lower the expression of TLR2 and TLR4 mRNA, the higher the expression of SIGIRR protein, the higher the proportion of G0/G1 phase cells, and the lower the proportion of S phase and G2 phase cells, and the higher the apoptosis rate(P<0.05). Conclusion: Aitongxiao prescription can alleviate pain in rats with liver cancer and promote cell apoptosis. The mechanism may be related to the regulation of TLR-NF-κB signaling.
