摘 要:目的:了解四川省乐山市围孕期女性细胞色素P4501A1基因(cytochrome P4501A1,CYP1A1)和谷胱甘肽硫转移酶基因M1(glutathione S-transferase M1,GSTM1)、T1(glutathione S-transferase T1,GSTT1)和P1(glutathione S-transferase P1,GSTP1)多态性分布特征。方法:选择2019年1月—2020年7月在乐山市人民医院进行围孕期保健的119名健康女性。采集口腔黏膜上皮脱落细胞,提取基因组DNA。使用荧光定量PCR方法检测CYP1A1、GSTM1、GSTT1和GSTP1基因多态性。结果:CYP1A1 Ile462Val基因位点野生纯合子AA基因型频率为54.6%,突变杂合子AG基因型频率为41.2%,突变纯合子G G基因型频率为4.2%,A等位基因频率为75.2%,G等位基因频率为24.8%;G S T P1 A313 G基因位点野生纯合子AA基因型频率为67.2%,突变杂合子AG基因型频率为31.1%,突变纯合子GG基因型频率为1.7%,A等位基因频率为82.8%,G等位基因频率为17.2%;GSTM1缺失基因型频率为58.0%,非缺失基因型频率为42.0%,GSTT1缺失基因型频率为43.7%,非缺失基因型频率为56.3%。结论:获取乐山市围孕期女性细胞色素P450基因和谷胱甘肽硫转移酶系相关基因多态性的群体遗传学特征,其中GSTM1和GSTT1相关基因位点缺失人群占比较高,这类人群对环境毒物的解毒能力低下,更应主动规避不良环境暴露,以及个体化营养干预来抵抗环境毒物带来的不良后果。Objective To understand the distribution characteristics of cytochrome P450 gene(CYP1A1)and glutathione sulfur transferase genes M1(GSTM1),T1(GSTT1)and P1(GSTP1)polymorphisms in women of childbearing age in Leshan City,Sichuan Province.Methods A total of 119 healthy women who received perinatal care in Leshan People’s Hospital from January 2019 to July 2020 were selected as the research subjects.The exfoliated epithelial cells of oral mucosa were collected to extract genomic DNA.CYP1A1,GSTM1,GSTT1 and GSTP1 gene polymorphisms were detected by fluorescence quantitative PCR.RESULTS:The frequency of wild homozygous AA genotype,mutant heterozygous AG genotype,mutant homozygous GG genotype was 54.6%,75.2%,and 24.8%in CYP1A1 ile462Val locus.The frequency of wild homozygous AA genotype,mutant heterozygous AG genotype,mutant heterozygous GG genotype,A allele and G allele were 67.2%,31.1%,1.7%,82.8%and 17.2%respectively.The frequency of GSTM1 genotype deletion was 58.0%,the frequency of non-deletion genotype was 42.0%,the frequency of GSTT1 genotype deletion was 43.7%,and the frequency of non-deletion genotype was 56.3%.Results The wild homozygous AA genotype frequency of CYP1A1 Ile462Val locus was 54.6%,the mutant heterozygous AG genotype frequency was 41.2%,the mutant homozygous GG genotype frequency was 4.2%,the A allele frequency was 75.2%,G etc.Allele frequency is 24.8%;Wild homozygous AA genotype frequency of GSTP1 A313G locus is 67.2%,mutant heterozygous AG genotype frequency is 31.1%,mutant homozygous GG genotype frequency is 1.7%,A allele frequency The frequency of G allele was 82.8%,the frequency of G allele was 17.2%;The frequency of missing genotypes in GSTM1 was 58.0%,the frequency of non-deletion genotypes was 42.0%,the frequency of missing genotypes in GSTT1 was 43.7%,and the frequency of non-deletion genotypes was 56.3.0%.Conclusion The population genetic characteristics of the cytochrome P450 gene and glutathione sulfur transferase related gene polymorphisms of women of childbearing age in Leshan City w
