摘 要:为了解猫杯状病毒(FCV)在中国华东地区的流行状况及其遗传变异和演化特征,本研究采集30份疑似感染FCV的猫眼鼻拭子,通过常规方法分离鉴定出9株FCV,经RT-PCR和测序获得了FCV VP1的基因序列,与文献报道的流行株、高致病力FCV(VS-FCV)株和疫苗株VP1基因同源性进行比对及遗传进化分析。结果显示,9株华东地区分离株VP1基因核苷酸序列同源性为70.9%~93.3%,分离株与流行株VP1基因核苷酸和氨基酸序列同源性分别为66.9%~84.8%和78.0%~91.9%,且大部分分离株与国内流行株处于同一分支;与VS-FCV VP1基因核苷酸和氨基酸序列同源性分别为73.9%~80.3%和82.2%~89.8%,其中只有FCV AH1分离株与VS-FCV-5株处于同一分支;与国内疫苗株VP1基因核苷酸和氨基酸序列同源性分别为73.9%~80.3%和82.2%~89.8%,其中只有FCV AH1株与国内疫苗株Vaccine-FCV-D90357处于同一分支。以上结果首次表明,华东地区FCV不断发生变异,是导致传统疫苗不能达到很好免疫保护效果的原因。本研究丰富了中国FCV的流行病学资料,为FCV疫苗研发提供了参考。In order to understand the prevalence of feline calicivirus(FCV)and its genetic variation and evolutionary direction in East China,30 conjunctival and nasal swabs were collected from FCV-suspected cats in this study.Nine FCV strains were isolated and identified by conventional methods,and VP1 genes of these FCV strains were obtained by RT-PCR and sequence analysis.Then,phylogenetic analyses based on the VP1 sequences were performed using the epidemic strains,the highly pathogenic FCV strain(VS-FCV)and the vaccine strain.The results showed that the nucleotide sequence similarity of these 9 FCV VP1 genes was 70.9%-93.3%,and the nucleotide and amino acid sequence similarities of the VP1 gene of these isolates and the epidemic strains were 66.9%-84.8%,and 78.0%-91.9%,respectively.Most of the isolates were in the same branch as the domestically epidemic strains.The nucleotide and amino acid sequence similarities with VS-FCV VP1 gene were 73.9%-80.3%and82.2%-89.8%,respectively.The FCV AH1 isolate was the only strain that located in the same branch as the VS-FCV-5 strain.The nucleotide and amino acid sequence similarities with the domestic vaccine strain VP1 gene were 73.9%-80.3%and 82.2%-89.8%,respectively.Among them,only the FCV AH1 strain was in the same branch as the domestic vaccine strain FCV-D90357.The data indicate that the continuous variation of FCV in East China is the reason why the traditional vaccines can not provide good immune protection.This study enriches the epidemiological data of FCV in China and provides a reference for the development of FCV vaccines.
