摘  要:通过在原始适体探针上添加自由辅助探针(assistant probe,AP)构建了一类基于置换反应信号机制的卡那霉素(kanamycin,KMN)适体传感器。该传感器以亚甲基蓝(methylene blue,MB)修饰的KMN适体为功能化捕获和信号探针(capturing and signaling probe,CSP),借助靶标与AP的竞争置换反应使适体探针发生构型变化而实现信号转导。实验对比考察了AP区域选择性对传感效率的影响,基础性考察了传感器的界面电化学行为和实际分析性能。结果表明,相比于单探针方式,背景电流明显减小,信号增长率(signal enhancement,SE)明显增大。将该传感器用于KMN检测,检测限为~0.3nmol/L,响应速度快,对于实际样品可在40min内完成全部检测过程。结合具有的其他优点如选择性好、抗污检测能力强和无需添加其他辅助检测试剂等,该传感器利于发展成为绿色化快速、灵敏的痕量KMN检测方法。A kanamycin(KMN)aptasensor based on the signaling mechanism of displacement hybridization reaction was constructed by adding a free assistant probe(AP)to the parent aptamer probe.Using a methylene blue(MB)-modified KMN aptamer as the capturing and signaling probe(CSP),the sensor can detect a conformation change in functional aptamer probe induced by the competitive displacement hybridization reaction between the target and the AP,thereby achieving the signal transduction.In addition to the contrastive investigation of the effect of AP regioselectivity on sensing efficiency,the interfacial electrochemical behavior and practical analytical perfo-rmance of the sensor were also investigated.The research results showed that a big increase in signal enhancement(SE)was achieved compared with the way of a single probe,which was helpful to obtain higher SE.When applied for the KMN detection,the proposed sensor highlighted a low detection limit of~0.3nmol/L and fast response speed with the whole detection process of<40min for the analysis of practical samples.Combined with other advantages of good selectivity,good anti-fouling detection ability,and reagentless detection process,this sensor is beneficial to being developed into a green rapid and sensitive detection method for the determination of trace KMN.
