摘 要:【目的】探讨天麻素对幼鼠癫痫的改善作用及对海马区星形胶质细胞增生的抑制作用。【方法】从60只SD幼鼠中随机选取50只注射氯化锂+匹罗卡品建立癫痫模型,剩余10只设为对照组(腹腔注射生理盐水)。将建模成功的44只幼鼠随机分为模型组(腹腔注射生理盐水)、茴香霉素组(腹腔注射5μg/kg茴香霉素)、天麻素+茴香霉素组(腹腔注射120 mg/kg天麻素+5μg/kg茴香霉素)、天麻素组(腹腔注射120 mg/kg天麻素),每组各11只。末次干预12 h后,监测脑电图观察脑电波改变情况,尼氏染色法观察海马神经元存活情况,末端脱氧核苷酸转移酶介导的d UTP缺口末端标记(TUNEL)染色法观察海马神经元凋亡情况,免疫组织化学法检测海马组织胶质纤维酸性蛋白(GFAP)表达情况,蛋白免疫印迹(Western Blot)法检测海马组织p38丝裂原活化蛋白激酶(MAPK)、磷酸化p38MAPK(p-p38MAPK)、B细胞淋巴瘤/白血病-2基因(Bcl-2)、Bcl-2相关X蛋白(Bax)蛋白表达情况。【结果】对照组脑电波以α、β波为主,偶有散在θ波;模型组出现以尖波、棘波、棘慢波为主的痫波;茴香霉素组痫波密度更大;天麻素+茴香霉素组、天麻素组痫波密度明显减小。与模型组比较,茴香霉素组潜伏期缩短,痫波频次和大发作次数增加,海马神经元存活细胞数减少、凋亡细胞数及GFAP表达阳性细胞数增加,p38MAPK、GFAP、Bax蛋白表达水平升高,Bcl-2蛋白表达水平降低(P<0.05);与茴香霉素组比较,天麻素+茴香霉素组、天麻素组潜伏期延长,痫波频次和大发作次数减少,海马神经元存活细胞数增加、凋亡细胞数及GFAP表阳性细胞数减少,p38MAPK、GFAP、Bax蛋白表达水平降低,Bcl-2蛋白表达水平升高(P<0.05);与天麻素+茴香霉素组比较,天麻素组潜伏期延长,痫波频次和大发作次数减少,海马神经元存活细胞数增加、凋亡细胞数及GFAP表阳性细胞数减少,p38MAPK、GFAP、Bax蛋白表达�Objective To explore the ameliorating effect of gastrodin on epilepyin young rats and its inhibitory effect on hyperplesia of astrocytes in hippocampus. Methods From 60 young SD rats,fifty were randomly selected and injected with lithium chloride + pilocarpine to establish epilepsy model. Forty-four successful modeling young rats were randomly divided into model group(intraperitoneal injection of normal saline), anisomycin group(intraperitoneal injection of 5 μg/kg of anisomycin), gastrodin +anisomycin group(intraperitoneal injection of120 mg/kg of gastrodin + 5 μg/kg of anisomycin), gastrodin group(intraperitoneal injection of 120 mg/kg of gastrodin),11 rats in each group,and the remaining 10 rats were set up as the control group(intraperitoneal injection of normal saline). After the last intervention for 12 hours,electroencephalo-graph(EEG)was monitored to observe the changes of brain waves,Nissl staining was used to observe the survival of hippocampal neurons,apoptosis of hippocampal neurons was observed by terminal deoxynucleotidyl transferase-mediated dUTP nick endlabeling(TUNEL)staining method, expression of glial fibrillary acidic protein(GFAP)in hippocampus was detected by immunohistochemistry, protein expression of p38 mitogen-activated protein kinase(p38 MAPK),phosphorylated p38 MAPK(p-p38 MAPK),B-cell lymphoma/leukemia-2 gene(Bcl-2)and Bcl-2 associated X protein(Bax)in hippocampus was detected by Western blotting assay. Results The brain waves in the control group were mainly α and β waves,with occasionally scattered θ waves. The epileptic waves in the model group were mainly sharp waves,spinous waves and spinous slow waves,and the density of epileptic waves in the anisomycin group was higher,the density of epileptic waves in the gastrodin+ anisomycin group and gastrodin group were more decreased significantly.
