摘 要:目的:建立HPLC-DAD波长切换联合梯度洗脱法同时测定10批新疆不同产地的一枝蒿药材中5种化学成分(异绿原酸A、异绿原酸C、蒙花苷、一枝蒿酮酸和金腰乙素)的分析方法。方法:采用HPLC-DAD法InertSustainR C18(250 mm×4.6 mm,5μm)色谱柱;乙腈(A)-0.2%磷酸(B)为流动相,流速为1.0 mL·min^(-1),梯度洗脱;异绿原酸A和异绿原酸C的检测波长为320 nm,蒙花苷检测波长为334 nm,一枝蒿酮酸检测波长为245 nm,金腰乙素检测波长为350 nm;进样量为10μL。建立一枝蒿药材指纹图谱,并进行相似度评价,采用SPSS 23.0统计软件对含量测定结果进行聚类分析。结果:10批样品指纹图谱有25个共有缝,相似度≥0.90;10批样品聚为2类。结论:该方法操作简便、快速、准确,为一枝蒿药材的质量控制及药用价值评价提供了科学依据。Objective:To develop an HPLC-DAD method for the simultaneous determination of the contents of 5 components(isochlorogenic acid A,isochlorogenic acid C,linarin,rupestonic acid,and chrysosptertin B)in Artemisia rupestris L.Methods:The chromatographic separation was achieved on a InerSustainR C18(250 mm×4.6 mm,5μm)column with acetonitrile(A)-0.2%phosphoric acid(B)as mobile phase at a flow rate of 1.0 mL·min^(-1) for gradient elution.Isochlorogenic acid A,isochlorogenic acid C,linarin,rupestonic acid and chrysosptertin B were detected at 320,320,334,245,350 nm,respectively.Injection volume was set at 10μL.To establish an HPLC chromatographic fingerprint of Artemisia rupestris L.,the similarity evaluation was carried out.The cluster analysis was performed by using SPSS 23.0 software.Results:There were 25 common peaks in HPLC fingerprint of 15 batches of Artemisia rupestris L.The similarities of 10 batches of samples were more than 0.85.The 10 batches of samples can be divided into 2 categories by cluster analysis.Conclusion:The method is simple,quick,accurate,and helpful for the quality control of Artemisia rupestris L.
