摘 要:目的:探讨丙泊酚对大鼠肠缺血-再灌注炎症反应的影响。方法:健康雄性SPF级SD大鼠40只,每组10只。采用夹闭肠系膜上动脉(SMA)1 h再灌注2 h方法制作肠缺血-再灌注损伤模型,随机分为假手术组(S组)、肠缺血-再灌注组(I/R组)、丙泊酚预处理组(P组)和生理盐水预处理组(NS组)。苏木素/伊红(HE)染色观察结肠组织的形态改变和病理损伤评分,酶联免疫吸附(ELISA)实验检测结肠组织白细胞介素-1(IL-1)、IL-6和肿瘤坏死因子-α(TNF-α)表达水平。结果:与S组比较,I/R组和NS组结肠组织损伤评分及IL-1、IL-6、TNF-α含量均增高(P<0.05)。与I/R组比较,P组结肠组织损伤评分及IL-1、IL-6、TNF-α含量均降低(P<0.05)。结论:丙泊酚腹腔注射预处理可以通过降低大鼠肠缺血-再灌注损伤程度及炎症反应。Objective:To investigate the effect of propofol on intestinal ischemia reperfusion inflammation in rats.Methods:40 healthy male SPF-grade SD rats were randomly divided into a sham operation group(group S),an intestinal ischemia reperfusion group(group I/R),a propofol pretreatment group(group P)and a normal saline pretreatment group(group NS),10 rats in each group.A model of ischemia-reperfusion injury was made by clamping the superior mesenteric artery(SMA)for 1 h and reperfusion for 2 h.Colon tissue was removed and stained with Hematoxylin/eosin(HE).Morphological changes of colon tissue were observed under light microscope,and pathological changes of colon tissue was performed.Enzyme linked immunosorbent assay(ELISA)test was used to detect colon tissue interleukin-1(IL-1),IL-6 and tumor necrosis factor-α(TNF-α).Results:Compared with group S,the score of colon tissue injury in group I/R and group NS was significantly increased(P<0.05),and contents of IL-1,IL-6 and TNF-αwere increased significantly(P<0.05).Compared with group I/R,the score of colon tissue injury in group P was decreased significantly(P<0.05),and the contents of IL-1,IL-6 and TNF-αwere decreased significantly(P<0.05).Conclusion:Propofol pretreatment by intraperitoneal injection can reduce intestinal ischemia-reperfusion injury by reducing the contents of IL-1,IL-6 and TNF-αin colon tissue.
