摘　　要：目的探讨抑癌基因T-cadherin在卵巢癌中的表达及其对卵巢癌细胞生物学行为的影响。方法从临床中收集45例卵巢癌和癌旁正常组织石蜡切片。采用免疫组化、逆转录荧光定量PCR以及Western blot分析T-cadherin在样本中的表达,采用相关性分析T-cadherin表达水平与临床特征之间的关联。培养卵巢癌细胞株A2780,实验分为未处理组和5-Aza-CdR组。分别采用RT-qPCR和Western blot分析T-cadherin表达变化。采用CCK-8分析细胞增殖。采用Transwell实验验证细胞侵袭能力。采用流式细胞术分析细胞凋亡。结果卵巢癌组织中T-cadherin的阳性率为33.3%(15/45),癌旁组织中T-cadherin的阳性率为86.7%(39/45),两组相比差异具有统计学意义(P<0.05)。T-cadherin表达与组织分化程度、FIGO分期之间密切相关(P<0.05)。与未处理组比较,5-Aza-CdR组中T-cadherin的表达水平显著上调(P<0.05)。T-cadherin过表达显著抑制细胞增殖和侵袭(P<0.05),促进细胞凋亡(P<0.05)。结论T-cadherin在卵巢癌组织中呈低表达,可能与卵巢癌恶性程度密切相关。激活T-cadherin可以抑制卵巢癌细胞的增殖、迁移,促进卵巢癌细胞凋亡。Objective To investigate the expression of tumor suppressor gene T-cadherin in ovarian cancer and its effect on the biological behaviors of ovarian cancer cells.Methods Immunohistochemistry,reverse transcription fluorescence quantitative PCR and Wes-tern blot were used to analyze the expression of T-cadherin in samples,and the correlation between T-cadherin expression level and the clinical pathological characteristics was analyzed.Ovarian cancer cell line A2780 was cultured and divided into untreated group and 5-AZA-CDR group.T-cadherin expression was analyzed by RT-qPCR and Western blot,respectively.Cell proliferation was analyzed by CCK-8.Cell invasion ability was verified by Transwell assay.Apoptosis was analyzed by flow cytometry.Results The positive rate of T-cadherin was 33.3%(15/45)in ovarian cancer tissues and 86.7%(39/45)in paracancerous tissues,and there was a statistically significant difference between the two groups(P<0.05).The expression of T-cadherin was closely correlated with degree of tissue differentiation and FIGO stage(P<0.05).5-Aza-CdR significantly up-regulated the expression of T-cadherin in A2780 cells(P<0.05).T-cadherin overexpression significantly inhibited the cell proliferation and invasion(P<0.05),and promoted the cell apoptosis(P<0.05).Conclusion The low expression of T-cadherin in ovarian cancer tissue may be closely related to the malignant degree of ovarian cancer.Activation of T-cadherin can inhibit the proliferation and migration of ovarian cancer cells and promote the apoptosis of ovarian cancer cells.
