利用丝线挂线造模法建立肛门直肠瘘大鼠模型及初步评价

摘　　要：目的:采用丝线挂线法建立肛门直肠瘘(简称肛瘘)大鼠模型,并对此模型进行初步的评价和验证。方法:将20只SD雄鼠适应喂养7d后随机分为正常组、模型组,每组10只,模型组采用大鼠肛周丝线挂线留置建立肛瘘模型,以一般状态、肛门温度、肛周局部分泌物变化、病理组织学观察作为初步评价指标。结果:与正常组比较,造模后模型组大鼠精神萎靡,活动度降低,肛温与肛周分泌物评分同步出现显著上升,第21天后呈逐渐下降趋势,肛周组织可见典型肛瘘病理表现:表层可见炎性肉芽组织与纤维结缔组织,肌层组织间伴炎细胞浸润。结论:本研究所采用的丝线挂线造模法可成功建立大鼠肛瘘模型,可为将来肛瘘的研究奠定基础。Objective:To establish an anal fistula model in rats by using silk-ligating method,to evaluate and validate the model preliminarily.Methods:Twenty male SD rats were adaptively raised for 7 days and randomly divided into the normal group and model group with 10 rats in each group.In the model group,perianal silk thread for ligation was indwelled.The general conditions,anal temperature,changes in perianal secretions,and pathological observation of perianal tissues were observe items.Results:The rats in the model group after modeling were listlessness and less active compared with the normal group.The average anal temperature and perianal secretion scores in the model group was higher than that of the normal group,it showed a downward trend after the 21th day.There were obvious pathological changes in the perianal tissues after sampling.Inflammatory granulation tissue and fibrosis could be seen on the surface,inflammatory cells with infiltrate on the muscularis tissue.Conclusion:In this study,the anal fistula model of rat is successfully established by using silk-ligating method.It will lay a foundation for the future study of anal fistula.
