摘  要:目的:观察健脾补肾、清肠化湿复方及其拆方对大鼠骨髓间充质干细胞(BMSCs)增殖趋化的影响及可能的作用机制,并探究其促增殖趋化最佳的功效药物配伍。方法:4~5周龄SD大鼠提取BMSCs,分别加入健脾补肾、清肠化湿复方及其拆方,CCK-8法检测BMSCs增殖情况,Transwell法检测BMSCs趋化能力,qRT-PCR法检测SDF-1和CXCR4基因表达水平,并以选择性CXCR4拮抗剂AMD3100为对照,进一步观察影响BMSCs增殖趋化的可能机制。结果:健脾补肾、清肠化湿方能够促进BMSCs增殖和趋化,拆方促增殖趋化作用依次为复方组>健脾清肠组>健脾补肾组>健脾组,且复方组能够上调趋化因子SDF-1和CXCR4基因表达,加入AMD3100后能够抑制促增殖趋化能力。结论:健脾补肾、清肠化湿方能够促进BMSCs的增殖和趋化,其机制可能与SDF-1/CXCR4信号轴有关。Objective:To investigate the effects and mechanism of Jianpi Bushen,Qingchang Huashi Compound and its decomposed prescriptions on the proliferation and chemotaxis capacity of BMSCs,and to explore the best drug compatibility for proliferation and chemotaxis.Methods:BMSCs were extracted from 4~5 weeks old SD rats,the subcultured BMSCs was added with Jianpi Bushen,Qingchang Huashi Compound and its decomposed prescriptions.CCK-8 was used to detect BMSCs proliferation,while Transwell chamber assay was used to detect BMSCs chemotactic ability.SDF-1 and CXCR4 gene expression levels was detected by qRT-PCR.The effect of the compound on the proliferation and chemotaxis of BMSCs was further observed with the selective CXCR4 antagonist(AMD3100)as the control.Results:Jianpi Bushen,Qingchang Huashi Compound significantly promoted the proliferation and chemotaxis of BMSCs,and up-regulated gene expression levels of SDF-1,CXCR4.The promotion effect of strengthening spleen had a better effect on proliferation and migration,the order was compound prescription group>Jianpi Qingchang group>Jianpi Bushen group>Jianpi group.The addition of AMD3100 inhibited the effect.Conclusion:Jianpi Bushen,Qingchang Huashi Compound and its decomposed prescriptions could significantly promote the proliferation and chemotaxis of BMSCs,its mechanism may be related to up-regulate the SDF-1/CXCR4 signals.
