摘 要:目的:探讨艾灸“天枢穴”对溃疡性结肠炎大鼠环状RNA(circRNA)表达谱的影响。方法:将雄性SD大鼠随机分为对照组、模型组、隔药灸组、西药组,每组8只,通过观察各组大鼠疾病活动度(DAI)、结肠宏观损伤评分、结肠组织病理学变化及评分(CMDI)明确艾灸对溃疡性结肠炎的效应。每组取3只大鼠结肠组织进行高通量测序,获取差异表达的circRNAs,并进行生物信息学分析。结果:与正常组比较,模型组溃疡形成,结肠黏膜上皮不完整,腺体异常,杯状细胞显著减少,可见大量炎性细胞浸润,DAI、结肠宏观损伤评分及CMDI评分均显著增高(P<0.01);结肠组织差异表达的circRNAs有99个。与模型组比较,隔药灸组和西药组大鼠DAI指数、结肠宏观损伤评分及CMDI评分均显著降低(P<0.01),差异表达的circRNAs分别有99、76个。艾灸干预前后差异表达的circRNAs亲本基因与免疫炎症密切相关。结论:艾灸“天枢穴”可能通过影响结肠组织circRNA表达谱,作用其亲本基因而调节免疫相关通路,达到减轻DSS诱导UC大鼠结肠炎症反应之目的。Objective:To explore the influence of herb-partitioned moxibustion at‘Tianshu(ST 25)’on the circRNA expression profiles in rats with ulcerative colitis(UC).Methods:Male SD rats were randomly divided into a normal group,model group,herb-partitioned moxibustion group and western medicine group,8 for each group.Disease activity index(DAI),and colonic mucosa damage index(CMDI)were observed to reveal the effect of moxibustion on UC.Total RNA was extracted from 3 rats in each group,and chose differentially expressed circRNAs by RNA-seq and bioinformatics.Results:Compared with the normal group,ulceration was formed the intestinal mucosa was not intact,the gland structure was destroyed,and there was a large amount of inflammatory cell infiltration in model group.UC rats exhibited colonic mucosa damage,with a reduced number of glands and substantial inflammatory cell infiltration,DAI and CMDI were significantly increased in the model group(P<0.01);there were 99 differentially expressed circRNAs.Compared with the model group,DAI and CMDI were significantly decreased in the herb-partitioned moxibustion group and the western medicine group(P<0.01).There were 99 and 76 differentially expressed circRNAs in the herb-partitioned moxibustion group and the western medicine group,respectively.The pathways of the parental genes of differentially expressed circRNAs in the herb-partitioned moxibustion group were closely related to immune inflammation and carcinogenesis.Conclusion:The moxibustion at‘Tianshu(ST 25)’may regulate the circRNA expression profiles,act on the parental genes to regulate inflammatory signaling pathways,and attenuate inflammation in the colons of rats with DSS-induced UC.
