摘 要:目的:探讨龟甲胶对环磷酰胺所致免疫低下型大鼠免疫调节作用。方法:从72只大鼠中随机抽取12只为空白组,剩余60只腹腔注射环磷酰胺制备免疫低下模型,并随机分为模型组,龟甲胶低、中、高剂量组及阳性组,每组12只。空白组连续灌胃0.9%氯化钠溶液14d,龟甲胶低、中、高剂量组分别按0.63、1.26、2.52g/kg灌胃龟甲胶溶液,阳性组按200mg/kg灌胃匹多莫德溶液,模型组给予等体积的0.9%氯化钠溶液灌胃,连续给药10d。给药结束后,观察大鼠免疫器官的脏器指数,HE染色进行病理组织形态学观察,流式细胞术检测外周血中T淋巴细胞计数,酶联免疫吸附试验(ELISA)检测血清中白介素-4(IL-4)、干扰素-γ(IFN-γ)的浓度,用荧光定量PCR法检测脾脏组织中IFN-γ、IL-4 mRNA的表达。结果:与空白组比较,模型组大鼠体质量、脾脏和胸腺指数显著下降(P<0.05,P<0.01),免疫器官出现病理形态学改变,外周血CD3^(+)、CD4^(+)T淋巴细胞计数显著降低(P<0.01),血清IFN-γ浓度显著降低(P<0.01),脾脏IFN-γmRNA表达显著降低,IL-4 mRNA表达显著上升,IFN-γ/IL-4显著降低(P<0.01)。与模型组比较,龟高组可提高大鼠体质量、脾脏及胸腺指数,改善免疫器官的病理损伤,促进淋巴细胞的增值分化,增加外周血中的CD3^(+)、CD4^(+)T淋巴细胞淋巴细胞计数,促进大鼠血清中IFN-γ浓度上升,脾脏的INF-γmRNA表达升高,差异均具有统计学意义(P<0.01)。结论:龟甲胶可以提高免疫功能低下大鼠的免疫功能,高剂量龟甲胶效果更显著。Objective:To investigate the immunomodulatory effect of tortoise shell glue on immunocompromised rats induced by cyclophosphamide.Methods:Twelve rats were randomly selected as blank group from 72 rats,and the remaining 60 rats were intraperitoneally injected with cyclophosphamide to prepare immunocompromised models,and were randomly divided into model group,tortoise shell gum low,medium and high dose groups and positive group,12 rats in each group.The blank group was administered with 0.9%sodium chloride solution for 14d,the tortoise shell gum solution in the low,medium,and highdose tortoise shell gum groups was administered with a concentration of 0.63,1.26 and 2.52g/kg,respectively.Pidotimod solution of 200mg/kg was intragastrically administered,and the model group was given an equal volume of normal saline intragastrically for 10 consecutive days.After the administration,the organ index and pathological tissue test(HE)of the rat immune organs were observed,flow cytometry was used to detect the T lymphocyte count in the peripheral blood,and enzyme-linked immunosorbent assay(ELISA)was used detect the serum interleukin-4(IL-4),the concentration of IL-4 and interferon-γ(IFN-γ)was used to detect the expression of IFN-γand IL-4 mRNA in the spleen tissue by fluorescence quantitative PCR.Results:Compared with the blank group,the body mass,the spleen and thymus index of the model group decreased(P<0.05,P<0.01),the immune organs showed pathological changes,and the CD3^(+)and CD4^(+)T lymphocyte counts in the peripheral blood decreased(P<0.01),the concentration of IFN-γin serum decreased(P<0.01),the expression of IFN-γmRNA in the spleen decreased,the expression of IL-4 mRNA increased,and IFN-γ/IL-4 decreased(P<0.01).Compared with the model group,the high-dose tortoise shell gum group can increase the body mass,the index of spleen and thymus,improve the pathological damage of immune organs,promote the proliferation and differentiation of lymphocytes,and increase the CD3^(+)and CD4^(+)T lymphocytes in the peripheral
