鳖甲煎丸联合骨髓间充质干细胞对大鼠肝纤维化的作用

摘　　要：目的:探讨鳖甲煎丸联合骨髓间充质干细胞(BMSC)移植疗法逆转大鼠肝纤维化效果。方法:通过从大鼠股骨干中分离提取干细胞,并利用其贴壁生长的特性进一步分离纯化出高纯度的BMSC,并分别采用流式细胞术、茜素红染色、油红“O”染色对分离纯化BMSC的纯度、成脂能力和成软骨能力对进行功能验证。造模成功的肝纤维化大鼠随机分为联合治疗组、干细胞治疗组、空白组3组,取正常大鼠为正常组。联合治疗组采用鳖甲煎丸灌胃联合BMSC尾静脉注射干预,干细胞治疗组单独采用BMSC尾静脉注射干预,空白组予0.9%氯化钠溶液干预,正常组为常规饲养大鼠。12周后进行肝功能检测与肝脏病理观察。结果:流式细胞术结果显示,分离纯化后的BMSCs的干细胞标志CD44阳性率达到91.46%,茜素红染色、油红“O”染色均呈阳性反应,联合治疗组在改善肝功能指标方面显著优于干细胞治疗组与空白组(P<0.05);干细胞治疗组在降低ALT显著优于空白组(P<0.05)。联合治疗组肝细胞纤维间隔消失,与正常组大鼠相比较无明显异常。结论:鳖甲煎丸联合BMSC对于改善肝纤维化大鼠肝功能及逆转肝纤维化状态具有一定的疗效,比单纯使用BMSC的疗效更佳。Objective:To investigate the effects of Biejiajian Pills combined with bone marrow mesenchymal stem cell(BMSC)transplantation on reversing liver fibrosis in rats.Methods:Stem cells were isolated from the femoral shaft of rats,and BMSCs with high purity were further isolated and purified by the characteristics of adherent growth.Flow cytometry,alizarin red staining and oil red‘O’staining were used to verify the purity,adipogenic capacity and chondrogenic capacity of BMSCs.The liver fibrosis rats were divided into three groups:combined treatment group,stem cell treatment group and blank group.The combined treatment group was given Biejiajian Pills by gavage combined with BMSC tail vein injection intervention;the stem cell treatment group was given BMSC tail vein injection intervention alone;the blank group was given 0.9%sodium chloride solution intervention;the normal group was given conventional feeding rats.After 12 weeks,liver function was detected and liver pathology was observed.Results:The results of flow cytometry showed that the positive rate of CD44 was 91.46%,and alizarin red staining and oil red‘O’staining were positive.Liver function index detection found that the combined treatment group was significantly better at improving liver function indexes than the stem cell treatment group and blank group(P<0.05);the stem cell treatment group was better than the control group in reducing ALT and other liver enzymes(P<0.05).The fibrous septum of hepatocytes disappeared in the combined treatment group,and there was no obvious abnormality compared with the normal group.Conclusion:Biejiajian Pills combined with BMSC has certain curative effect on improving liver function and reversing liver fibrosis in rats with liver fibrosis,which is better than BMSC alone.
