摘 要:目的:探讨灸法对逼尿肌反射亢进大鼠P2X3受体表达和传入神经C神经纤维的影响。方法:健康成年雌性SD大鼠随机分为假手术组、模型组、艾灸组,每组6只。采用T9完全脊髓横断方法建立逼尿肌反射亢进大鼠模型。采用荧光定量PCR方法和Western Blot检测膀胱黏膜P2X3受体mRNA和蛋白表达,采用荧光素-荧光素酶法检测膀胱黏膜ATP含量,采用硝酸还原法检测膀胱黏膜NO含量;采用电生理信号采集系统分析灸法对C神经纤维的影响。结果:与假手术组比较,模型组大鼠盆神经放电次数增加(P<0.01),膀胱黏膜P2X3受体mRNA和蛋白表达显著增加(P<0.01),膀胱黏膜ATP含量上升(P<0.01),NO含量下降(P<0.01);与模型组比较,艾灸组大鼠膀胱黏膜P2X3受体mRNA和蛋白表达显著减少(P<0.01),传入神经兴奋性均显著减弱(P<0.01),膀胱黏膜ATP含量显著下降(P<0.01),NO含量显著上升(P<0.01)。结论:灸法可以降低逼尿肌反射亢进大鼠膀胱黏膜P2X3受体mRNA和蛋白表达,使膀胱黏膜ATP含量下降,NO含量上升,可以降低传入神经C神经纤维的过度活动。Objective:To study the effects of moxibustion on P2X3 receptor expression and afferent C nerve fiber in rats with detrusor hyperreflexia.Methods:Healthy adult female SD rats were randomly divided into sham operation group,model group,and moxibustion group,with 6 rats in each group.The rat model of detrusor hyperreflexia was established by T9 complete spinal cord transection.The expression of P2X3 receptor mRNA and protein in bladder mucosa was detected by fluorescence quantitative PCR and Western Blot.The content of ATP in bladder mucosa was detected by fluorescein-luciferase method,and the content of NO in bladder mucosa was detected by nitrate reduction method.The effect of moxibustion on C nerve fiber was analyzed by electrophysiological signal acquisition system.Results:Compared with the sham operation group,the discharge times of pelvic nerve increased in the model group(P<0.01),the expression of P2X3 receptor mRNA and protein was significantly increased in the model group(P<0.01),the content of ATP in the bladder mucosa increased(P<0.01),the content of NO decreased(P<0.01).Compared with the model group,the expression of P2X3 receptor mRNA and protein in bladder mucosa of moxibustion group was significantly decreased(P<0.01),the excitability of afferent nerve was significantly decreased(P<0.01),the content of ATP in bladder mucosa was significantly decreased(P<0.01),and the content of NO was significantly increased(P<0.01).Conclusion:Moxibustion could reduce the expression of P2X3 receptor mRNA and protein in bladder mucosa of rats with detrusor hyperreflexia,reduce the content of ATP and increase the content of NO in bladder mucosa,and reduce the hyperactivity of afferent C nerve fibers.
