摘 要:目的:研究慈姑多糖(SSP)对异烟肼(INH)和利福平(RFP)联用致HepG2细胞损伤中血红素加氧酶-1(HO-1)和谷氨酰-L-半胱氨酸连接酶催化亚单位(GCLC)酶含量、基因及蛋白表达影响。方法:采用ELISA检测HepG2细胞中HO-1和GCLC含量变化;采用定量RT-PCR技术检测HepG2细胞中HO-1、GCLC mRNA的表达;采用Western Blot法检测HepG2细胞中HO-1和GCLC蛋白的表达。结果:与正常对照组比较,模型组HepG2细胞中HO-1和GCLC含量、mRNA及蛋白表达显著增加(P<0.05);与模型组比较,SSP最佳给药组中HO-1和GCLC mRNA、蛋白表达显著升高(P<0.05)。结论:SSP可能通过促进HepG2细胞中HO-1及GCLC含量、mRNA及蛋白的表达,发挥其对INH/RFP联用致肝损伤的保护作用。Objective:To study the effects of Sagittaria Sagittifolin polysaccharide(SSP)on the content of HO-1 and GCLC enzymes,genes and proteins expression in HepG2 cell injury caused by the combined use of isoniazid and rifampicin(INH/RFP).Methods:Enzyme-linked immunoassay(ELISA)was used to detect the changes of HO-1 and GCLC content in HepG2 cells;quantitative RT-PCR was used to detect the expression of HO-1 and GCLC content mRNA in HepG2 cells;Western Blot method was used to detect HepG2 cells HO-1 and GCLC protein expression.Results:Compared with the normal control group,the model group could increase the HO-1 and GCLC content,mRNA and protein expression in HepG2 cells(P<0.05).Compared with the model group,the mRNA and protein expression of HO-1 and GCLC in the SSP best administration group was significantly increased(P<0.05).Conclusion:SSP may play a protective role in the liver injury induced by the INH/RFP through promoting the content of HO-1 and GCLC mRNA and protein expression in HepG2 cells.
