摘 要:目的:研究复方七芍降压片对自发性高血压(SHR)大鼠肾动脉中G蛋白偶联受体P2Y6、磷酯酶C(PLC)、核因子κB抑制蛋白(I-κB)、核因子κB(NF-κB)蛋白表达的影响,探讨复方七芍降压片干预血管重塑的作用机制。方法:SPF级雄性SHR大鼠30只随机分为模型组、阳性药组(13.5mg/kg厄贝沙坦片)、复方七芍降压片组(8.73g/kg复方七芍降压片),另设10只WKY大鼠为正常对照组。各组分别给予相应处理6周后取材,HE染色法观察血管形态;ELISA法检测血清中肿瘤坏死因子-α(TNF-α)水平;免疫组化法检测肾动脉中P2Y6、PLC、I-κB、NF-κB的蛋白表达水平。结果:与正常对照组比较,模型组大鼠动脉内膜粗糙不平,甚至有内膜脱落情况,平滑肌细胞数量增加,分布不均匀,排列紊乱,血清TNF-α水平及肾动脉中P2Y6、PLC、I-κB、NF-κB蛋白表达水平显著升高(P<0.01,P<0.05)。药物干预后,与模型组比较,复方七芍降压片组内膜平整,无脱落情况,平滑肌细胞数量无明显增加,分布均匀,排列大致整齐,血清TNF-α水平及肾动脉中P2Y6、PLC、I-κB、NF-κB蛋白表达水平显著下降(P<0.05)。结论:复方七芍降压片能够显著改善血管重塑;其机制与抑制TNF-α的分泌、调控肾动脉中P2Y6/PLC/I-κB/NF-κB轴有关。Objective:To study the effects of compound Qishao Jiangya Tablet on P2Y_(6)/PLC/I-κB/NF-κB axis in renal artery of spontaneously hypertensive rats(SHRs),and further to explore the mechanism of intervention of compound Qishao Jiangya Tablet on vascular remodeling.Methods:A total of 30 SPF male SHRs were randomly divided into model group,irbesartan tablet group(13.5mg/kg),compound Qishao Jiangya Tablet group(8.73g/kg),and another 10 WKY rats as normal control group.After six weeks of treatment,the blood vessels of each group were observed by HE staining,the level of TNF-αin serum was detected by ELISA,and the protein expression levels of P2Y_(6),PLC,I-κB and NF-κB in renal artery were detected by immunohistochemistry.Results:Compared with the normal control group,the intima of the model group was rough and uneven,and even the intima was exfoliated,the number of smooth muscle cells increased,the distribution was uneven,the arrangement was disordered,the level of TNF-αin serum and t he expression level of P 2Y_(6),PLC,I-κB and NF-κB in renal artery increased significantly(P<0.01,P<0.05).After drug intervention,compared with the model group,the inner membrane of the compound Qishao Jiangya Tablet group was smooth without shedding,the number of smooth muscle cells was not significantly increased,the distribution was uniform,the arrangement was roughly orderly,the level of serum TNF-αand the expression of P2Y_(6),PLC,I-κB and NF-κB protein in the renal artery were significantly decreased(P<0.05).Conclusion:Compound Qishao Jiangya Tablet can significantly improve vascular remodeling,and its mechanism is related to the inhibition of TNF-αsecretion and the regulation of P2Y6/PLC/I-κB/NF-κB axis in renal artery.
