摘  要:目的:研究五加减正气散(WJJZQS)含药血清对To ll样受体3(TLR3)信号通路及相关炎症因子表达的影响。方法:制备染毒细胞模型(RV SA11株,MA104细胞)。Wistar大鼠按体质量随机分为空白组,中成药组,西药组及WJJZQS大、中、小剂量组,按剂量灌胃相应药物,2次/d,连续给药3d,于末次灌胃1h后眼眶取血。检测各含药血清作用染毒细胞后,TLR3信号通路及相关炎症因子的表达。结果:与染毒组比较,WJJZQS大、中剂量组能显著下调染毒细胞白介素(IL)-6、IL-1β、肿瘤坏死因子-α(TNF-α)的高表达,上调干扰素β(IFN-β)的低表达(P<0.05,P<0.01);WJJZQS小剂量组对IL-1β、TNF-α也有一定的调控作用(P<0.05);WJJZQS大、中剂量组可显著下调TLR3、TRIF mRNA及蛋白的过表达(P<0.01,P<0.05),但对IRF3 mRNA及蛋白调控作用不明显。结论:WJJZQS含药血清可通过调控TLR3信号通路关键蛋白及下游炎症因子途径发挥一定的抗病毒作用。Objective:To study the effects of the serum containing Wujiajian Zhengqi Powder(WJJZQS)on TLR3 signaling pathway and the expression of related inflammatory factors.Methods:Preparation of infected cell model(RV SA11 strain,MA104 cells).Wistar rats were randomly divided into 6 groups according to body weight:blank group,Chinese patent medicine group,Western medicine group and WJJZQS large,medium and small dose groups.Gavage the appropriate drugs according to the dose,twice a day.The drug was administered continuously for 3 days,and blood was collected from the eye socket after 1h of the last gavage.The expression of TLR3 signaling pathway and related inflammatory factors were detected after each medicated serum acted on infected cells.Results:Compared with the infected group,WJJZQS large and medium dose groups could significantly down-regulate the high expression of IL-6,IL-1βand TNF-α,up-regulated the low expression of IFN-βin infected cells(P<0.05,P<0.01).The small dose group also had certain regulatory effects on IL-1βand TNF-α(P<0.05).WJJZQS large and medium dose groups could significantly down-regulate the high expression of TLR3,TRIF mRNA and overexpression of protein(P<0.05,P<0.01).However,IRF3 mRNA and protein were not significantly regulated.Conclusion:The serum containing WJJZQS can play a certain antiviral effect by regulating the key proteins of TLR3 signaling pathway and downstream inflammatory factor pathway.
