摘  要:目的:研究细辛醇提物对热敏通道瞬时感受器电位香草素受体1(TRPV1)的干预作用,探讨细辛止痛的分子机制。方法:运用全细胞膜片钳技术测定细辛醇提物对hTRPV1/HEK293细胞跨膜电流的影响。小鼠足底注射福尔马林诱发疼痛,观察细辛是否具有止痛作用。结果:与细胞外液比较,3、10mg/mL细辛醇提物能使hTRPV1/HEK293细胞产生显著的跨膜电流(P<0.05,P<0.01)。10mg/mL细辛醇提物诱导电流显著大于3mg/mL(P<0.01)。福尔马林诱导的疼痛第一阶段(0~5min)和第二阶段(10~30min),与对照组比较,模型组小鼠抬足频数显著增加及抬足时间延长(P<0.01);阳性药组和细辛高、中剂量组能显著减少小鼠的抬足频数并缩短抬足时间(P<0.05,P<0.01),且在福尔马林诱导的疼痛第二阶段(10~30min),细辛低剂量组能显著缩短小鼠抬足时间(P<0.01)。结论:细辛醇提物中含有热敏通道TRPV1的激动剂,细辛能减轻福尔马林诱导的疼痛反应,中药细辛止痛机制可能与热敏通道TRPV1被激活有关。Objective:To study the effects of ethanol extract of Asari Radix on the thermosensitive channel TRPV1,and to explore the molecular mechanism of the analgesic effect of Asari Radix.Methods:The effects of Asari Radix extract on the transmembrane current of hTRPV1/HEK293 cells was determined by the whole-cell patch clamp technique.To observe whether Asari Radix has analgesic effect by formalin test.Results:Compared with extracellular fluid,the 3mg/mL and 10mg/mL Asari Radix ethanol extract enabled hTRPV1/HEK293 cells to produce a significant transmembrane current(P<0.05,P<0.01).The induced current of 10mg/mL Asari Radix ethanol extract was significantly greater than 3mg/mL Asari Radix ethanol extract(P<0.01).In the first phase of formalin-induced pain(0~5min)and the second phase(10~30min),compared with the control group,the model group showed a significant increase in the frequency and the duration of foot lifting(P<0.01);Ibuprofen,high and medium doses of Asari Radix significantly reduced the frequency of foot lifting and shortened the time of foot lifting in mice(P<0.05,P<0.01).In the second phase(10~30min)of formalin-induced pain,compared with the control group,the low doses of Asari Radix significantly shortened the time of lifting in mice(P<0.01).Conclusion:Asari Radix ethanol extract contains the agonist of the thermosensitive channel TRPV1,which can alleviate the formalin-induced pain response,and the mechanism of analgesia of Asari Radix may be related to the activation of the thermosensitive channel TRPV1.
