摘  要:目的:探究益气温阳护卫法防治支气管哮喘的作用机制。方法:建立动物哮喘模型,造模大鼠分别于第1天腹腔注射新鲜配制的卵蛋白氢氧化铝溶液1mL(内含卵蛋白10mg及氢氧化铝200mg)致敏,并于第8天重复注射1次。第15天起予以2%卵蛋白雾化吸入激发哮喘,1次/d,30min/次,分别持续1、4周。第16天(即致敏后)开始,每天雾化吸入卵蛋白前1h,按0.36、0.72、1.44g/kg灌服益气温阳护卫汤药液。大鼠从雾化吸入2%OVA起,分别于激发1、4周处死大鼠各6只,各组大鼠于末次雾化吸入后24h内用1%戊巴比妥钠腹腔注射麻醉后,将其固定于手术台上,分离腹主动脉,取动脉血,ELISA检测白介素-1β(IL-1β)表达;肺组织以液氮冻存,HE染色观察组织形态学改变,qPCR检测人防御素-5(HD5)的基因表达。结果:HE染色显示,益气温阳护卫高剂量组管壁增厚有明显改善作用。与对照组比较,第1、4周模型组IL-1β含量均显著升高(P<0.05);与模型组比较,第1周益气温阳护卫中、高剂量组与第4周益气温阳护卫各剂量组IL-1β含量均显著降低(P<0.05)。与对照组比较,第1、4周模型组HD5基因表达均显著升高(P<0.05);与模型组比较,益气温阳护卫各剂量组HD5表达均显著降低(P<0.05)。结论:益气温阳护卫法防治支气管哮喘的作用机制可能是通过调节防御素的表达实现。Objective:To explore the mechanism of Yiqi Wenyang Huwei method for the prevention and treatment of bronchial asthma.Methods:An animal asthma model was established.On the first day,rats in each model group were injected intraperitoneally with freshly prepared ovalbumin(OVA)aluminum hydroxide solution 1mL(containing OVA 10mg and aluminum hydroxide 200mg)to sensitize them.Repeat the injection once on 8th day.Inhale 2%OVA from day 15 to stimulate asthma,once a day,30min each time,lasting 1 to 4 weeks.Beginning on the 16th day(that was,after sensitization),1h before inhalation of OVA by inhalation every day,administer Yiqi Wenyang Huwei Decoction at 0.36,0.72,1.44g/kg.The rats were sacrificed by inhaling 2%OVA,and 6 rats were sacrificed at 1 week and 4 weeks after challenge,respectively.Rats in each group used 1%pentobarbital within 24h after the last inhalation Sodium intraperitoneally.After anesthesia,it was fixed to operating table,the abdominal aorta was isolated,arterial blood was taken,and the expression of IL-1βwas detected by ELISA;lung tissue was frozen in liquid nitrogen,HE staining was used to observe histomorphological changes,and qPCR was used to detect HD5 expression.Results:HE staining results showed that the high dose group of Yiqi Wenyang Huwei had a significant improvement in the role of thickening significantly weakened.Compared with the blank control group,the expression of IL-1βin the model group in the first and fourth weeks was higher(P<0.05).Compared with the model group,the expression of IL-1βin the middle-and high-dose groups of Yiqi Wenyang Huwei in the first week and the low-,medium-,and high-dose groups of Yiqi Wenyang Huwei in the fourth week were lower(P<0.05).Compared with the blank control group,the expression of HD5 gene in the model group was higher in the first and the fourth week(P<0.05).Compared with the model group,the HD5 expression in all doses groups of Yiqi Wenyang Huwei were lower(P<0.05).Conclusion:The mechanism of prevention and treatment of bronchial asthma with the m
