温肾复方对冰羟基脲建立的肾阳虚证不孕小鼠生殖功能影响

摘　　要：目的:探讨温肾复方中药对肾阳虚证不孕小鼠生殖功能的影响。方法:将4周龄昆明雌性小鼠随机分为对照组、模型组、温肾复方组。按冰羟基脲灌胃法建立模型后,对照组和模型组予0.9%氯化钠溶液,温肾复方组予温肾复方灌胃。观察各组小鼠一般情况、肛温,药物干预后各组小鼠合笼观察子代,用PMSG 5IU+HCG 5IU进行超促排卵获取卵母细胞进行分期计数,检测血清雌二醇(E2)、孕激素(P)、抗缪勒管激素(AMH)水平,运用HE染色观察卵巢病理改变。结果:与对照组比较,模型组小鼠产仔数、获卵数、E2、P及AMH水平、卵泡期及黄体期原始卵泡数均显著下降(P<0.01,P<0.05);异卵率、卵泡期及黄体期闭锁卵泡数显著增加(P<0.01)。与模型组比较,温肾复方组小鼠产仔数、获卵数、卵泡期E2、AMH水平、卵泡期原始卵泡数显著增加(P<0.01,P<0.05);异卵率、卵泡期及黄体期闭锁卵泡数显著下降(P<0.01,P<0.05)。结论:温肾复方中药能改善肾阳虚不孕小鼠生殖功能,与正向调节性腺和下丘脑-垂体-卵巢(HPO)轴功能有关。Objective:To explore the effect of traditional Chinese medicine of Wenshen Compound on reproductive function of infertile mice with kidney yang deficiency syndrome.Methods:4-week-old Kunming female mice were randomly divided into control group,model group and Wenshen Compound group.After the model was established by intragastric administration of ice hydroxyurea,0.9%sodium chloride solution was given to the control group and model group,and Wenshen Compound was given to the Wenshen Compound group.The general condition and anal temperature of mice in each group were observed.After drug intervention,the offspring were observed in cages.The oocytes were obtained by superovulation with PMSG 5IU+HCG 5IU and counted by stages.The levels of serum estradiol(E2),progesterone(P)and anti-Müllerian hormone(AMH)were detected,and the pathological changes of ovary were observed by HE staining.Results:Compared with the control group,the litter size,the number of oocytes obtained,the levels of E2,P and AMH,the number of primordial follicles in follicular phase and luteal phase in the model group decreased significantly(P<0.01,P<0.05),while the rate of heterozygote and the number of atretic follicles in follicular phase and luteal phase increased significantly(P<0.01).Compared with the model group,the litter size,the number of oocytes obtained,the level of E2 and AMH in follicular phase and the number of primordial follicles in follicular phase increased significantly in Wenshen Compound group(P<0.01,P<0.05),while the number of atretic follicles in follicular phase and luteal phase decreased significantly in Wenshen Compound group(P<0.01,P<0.05).Conclusion:Wenshen Compound can improve the reproductive function of infertile mice with kidney yang deficiency syndrome,which is related to the positive regulation of gonad and hypothalamus-pituitary-ovary(HPO)axis.
