摘  要:目的研究黑地黄丸对肾性骨病模型大鼠改善作用机制。方法按照体重将Wistar大鼠随机分为6组:正常组、模型组、对照组(灌胃骨化三醇4.5×10-2μg·kg^(-1))和低、中、高3个剂量实验组(按10 mL·kg^(-1)药液灌胃黑地黄丸0.5,1.0,2.0 g·mL^(-1)),每组10只;1次/天,连续60 d。除正常组外,其余各组大鼠均行5/6肾切除并给予高磷饮食建立肾性骨病模型。Micro-CT扫描检测大鼠股骨骨密度(BMD)值和骨体积分数(BV/TV)百分率,以蛋白质印迹法检测骨组织骨形成蛋白-7(BMP-7)的表达。结果正常组、模型组、对照组和高剂量实验组的BMD值分别为(195.23±18.46),(137.33±16.12),(178.24±9.48)和(175.33±9.75)mg HA·ccm^(-1);这4组的骨体积分数百分率分别为(0.53±0.06)%,(0.32±0.06)%,(0.47±0.06)%和(0.45±0.06)%;这4组的BMP-7蛋白表达水平分别为0.72±0.02,0.32±0.02,0.67±0.02和0.68±0.04。上述指标:模型组与正常组比较,或对照组和高剂量实验组与模型组比较,差异均有统计学意义(均P<0.01)。结论黑地黄丸通过调控骨组织BMP-7蛋白表达,改善钙磷代谢、增加骨密度值与骨体积分数,有效改善大鼠的肾性骨病。Objective To study the effect and mechanism of Heidihuang Pills on model rats with renal osteopathy.Methods Wistar rats were randomly divided into six groups according to weight:normal group,model group,control group(was given 4.5×10-2μg·kg^(-1)calcitriol)and low,medium,high dose experimental groups(was given Heidihuang pills of 10 mL·kg^(-1)with 0.5,1.0,2.0 g·mL^(-1)),with 10 rats in each group;once a day for 60 d.Except the normal group,5/6 nephrectomy and high phosphorus diet were used to establish renal osteopathy model.The bone mineral density(BMD)and percentage of bone volume fraction(BV/TV)of femur were measured by micro CT scanning.The expression of bone morphogenetic protein-7(BMP-7)protein was detected by Western blot.Results The BMD values of normal group,model group,control group and high dose experimental group were(195.23±18.46),(137.33±16.12),(178.24±9.48)and(175.33±9.75)mg HA·ccm^(-1);the percentage of BV/TV in the 4 groups were(0.53±0.06)%,(0.32±0.06)%,(0.47±0.06)%and(0.45±0.06)%;the expression of BMP-7 protein in the 4 groups were 0.72±0.02,0.32±0.02,0.67±0.02 and 0.68±0.04.Comparison between model group and normal group,or control group,high dose experimental group and model group,the differences of the factors were significant(all P<0.01).Conclusion Heidihuang pills can effectively improve the renal bone disease in rats by regulating the expression of BMP-7 protein in bone tissue,improving calcium and phosphorus metabolism,increasing bone density and bone volume fraction.
