摘 要:目的探讨小鼠胚胎心包内动脉心外膜的起源、形成及其所含细胞在心包内主、肺动脉管壁发育过程中可能的作用。方法取胚龄E9.5—E16的小鼠胚胎石蜡包埋连续切片,进行免疫组织化学染色。提取及培养E13小鼠胚胎心包内主、肺动脉组织原代心外膜祖细胞,进行免疫荧光染色。结果E9.5—E10.5小鼠胚胎,心外膜逐渐形成并分布于心房、房室管和心室壁外表面,但流出道壁心外膜尚未形成;E11—E11.5小鼠胚胎,流出道非心肌部逐渐分隔为心包内升主动脉和肺动脉干,动脉心外膜开始出现在心包腔背侧壁以及与之相连的流出道壁远端;E12.5—E16小鼠胚胎,心包内主、肺动脉完全分隔,第二生心区前体细胞与动脉心外膜向平滑肌细胞分化,动脉瓣膜内也可见动脉心外膜来源的心外膜细胞。结论小鼠胚胎流出道非心肌部心外膜来自于动脉端心包腔背侧壁脏壁中胚层,动脉心外膜来源的间充质细胞可以进入动脉中膜,参与动脉壁的发育,同时参与动脉瓣膜的发育。Objective To investigate the origin and formation of intrapericardial arterial epicardium(aEP)and the possible role of its cells in the development of intrapericardial aorta and pulmonary artery walls in mouse embryos.Methods The paraffin-embedded serial sections of E9.5-E16 mouse embryos were stained with immunohistochemistry and immunofluorescence.The primary epicardial progenitor cells from the intrapericardial aorta and pulmonary artery tissue of E13 mouse embryos were extracted and cultured for immunofluorescence staining.Results In E9.5 to E10.5 mouse embryos,the epicardium gradually formed and distributed on the outer surface of the atrium,atrioventricular canal and ventricular wall,but the epicardium of the outflow tract(OFT)wall had not yet formed.In E11 to E11.5 mouse embryos,the non-myocardial part of the OFT was gradually separated into the intrapericardial ascending aorta and pulmonary artery trunk,and the aEP began to appear in the dorsal wall of the pericardial cavity and the distal end of the OFT wall connected to it.During E12.5 to E16,the aorta and pulmonary artery were completely separated.Progenitor cells of the second heart field and aEP differentiated into smooth muscle cells,and epicardial cells derived from the aEP can also be seen in the arterial valve.Conclusion The epicardium of the non-myocardial part of mouse embryonic OFT comes from the visceral mesoderm in the dorsal wall of artery-end pericardial cavities.Mesenchymal cells derived from the aEP can enter the tunica media of the arteries and participate in the development of arterial walls and valves at the same time.
