摘 要:目的:在经典方剂知柏地黄丸治疗肾阴虚证确有疗效的前提下,寻找知柏地黄丸在肾阴虚治疗上的质量标志物。方法:拟以中医方证代谢组学为核心策略,联合高通量UPLC-MS技术,寻找肾阴虚证的血液代谢标记物,在与显效状态下的知柏地黄丸血中移行成分建立相关性的前提下,揭示知柏地黄丸的药效物质基础,确定知柏地黄丸的质量标志物。结果:鉴定肾阴虚证的26个血液代谢标记物,其中19个代谢标记物在治疗后被知柏地黄丸显著调节,共鉴定显效状态下的47个血中移行成分,其中11个成分被鉴定为知柏地黄丸治疗肾阴虚证的药效物质基础。结论:根据中药质量标志物的相关原则,推测丹皮酚、没食子酸、棉子糖、芍药苷、药根碱、泽泻醇B-单乙酸酯等6种体内原型成分为知柏地黄丸治疗肾阴虚证的质量标志物。Objective:To trace the quality marker of the classic formula Zhibai Dihuang Pill(ZBDH),on the premises it is effective in the treatment of Kidney-yin deficiency.A high-throughput UPLC-MS based on Chinmedomics strategy was applied to establish the correlation between active ingredients and biomarkers to discover the material basis of ZBDH,to reveal its combination advantages and to identify the potential quality markers(Q-markers)under its therapeutic efficacy.Results:In this study,26 potential biomarkers had been identified,19 biomarkers were significantly modulated after the treatment by ZBDH,47 blood-migrated components had been identified and 11 components were identified as the effective substance basis of ZBDH.Conclusion:According to the principles of Q-markers of Chinese medicinal materials,six original components were speculated as Q-markers of ZBDH,which were paeonol,gallic acid,raffinose,paeoniflorin,jatrorrhizine and alisol B monoacetate.
