中药贴膜对兔膝骨性关节炎关节软骨下骨BMP-2蛋白表达的影响

摘　　要：目的:通过研究中药贴膜对早期兔膝关节骨性关节炎模型关节软骨下骨内骨形态发生蛋白-2(BMP-2)表达的影响,分析评价其治疗骨性关节炎作用机理。方法:应用改良的Hulht方法建立兔右膝关节骨性关节炎模型,随机分为模型组、传统中药组、玻璃酸钠组、中药贴膜组。分别采用医用橡皮膏外敷、传统方法粉碎的中药外敷、玻璃酸钠关节内注射、中药贴膜外敷治疗,另取10只健康白兔作为空白对照组。治疗6周后通过免疫组化和Western blot方法测定各组实验动物右膝关节软骨下骨中BMP-2表达的变化。结果:免疫组化结果和Western blot结果均显示,各组动物经过治疗后其右膝关节软骨下骨中BMP-2表达有不同变化,其中空白对照组最高,模型组最低,玻璃酸钠组高于模型组,但低于传统中药组和中药贴膜组,中药贴膜组BMP-2表达量与空白对照组接近。结论:中药贴膜可以上调兔骨性关节炎模型关节软骨下骨中BMP-2表达,且疗效优于玻璃酸钠组和传统中药组,说明中药贴膜治疗骨性关节炎作用机理与调节关节软骨下骨BMP-2表达量有关。Objective:To study the effect of the application of Chinese medicine film on the expression of bone morphogenetic protein-2(BMP-2)in subchondral bone of the rabbits with early knee osteoarthritis(KOA),thus to analyze its mechanism in treating osteoarthritis(OA).Methods:The OA model of right knee was established by modified Hulht method,and the successful models were randomly divided into the model group,the traditional Chinese medicine group,the Sodium Hyaluronate group and the application of Chinese medicine film group,which were treated with external application of medical plaster,external application of traditional Chinese medicine powder,intra-articular injection of Sodium Hyaluronate and external application of Chinese medicine film respectively.Another 10 healthy rabbits were taken as the blank control.After 6 weeks of the treatment,the expression of BMP-2 in subchondral bone of knee joint was detected by immunohistochemistry and Western blot methods.Results:The results of immunohistochemistry and Western blot showed that the expression of BMP-2 in the subchondral bone of each group had different changes after the treatment,among which the blank control group was the highest;the model group was the lowest;the Sodium Hyaluronate group was higher than the model group,but lower than the traditional Chinese medicine group and the application of Chinese medicine film group;the expression of BMP-2 in the application of Chinese medicine film group was close to that in the blank control group.Conclusion:The application of Chinese medicine film can up-regulate the expression of BMP-2 in articular subchondral bone,and it is better than Sodium Hyaluronate and the application of traditional Chinese medicine.It shows that the mechanism of the application of Chinese medicine film is related to the regulation of BMP-2 expression in subchondral bone in the treatment of OA.
