摘  要:为了解湿地微生物的组成和功能,合理利用和开发湿地微生物资源,采用Illumina高通量测序平台对天津北大港湿地沉积物的细菌16S rRNA基因(V4-V5区)进行高通量测序,检测0~8、8~22和22~28 cm沉积物中的细菌群落结构和多样性.结果表明:从表层到底层获得的细菌OTUs分别为1716、2000和1263个.明确分类地位的细菌有16个门42纲70个目116科187属.在0~8 cm沉积物中,变形菌门(44.73%)、绿弯菌门(16.84%)和酸杆菌门(10.25%)是优势菌群;在8~22 cm沉积物中,变形菌门、绿弯菌门和酸杆菌门仍然是优势菌群,相对丰度分别为37.24%、14.02%和11.27%;在22~28 cm沉积物中,厚壁菌门、变形菌门和绿弯菌门是优势类群,相对丰度分别为45.84%、21.95%和6.26%.0~22 cm沉积物细菌菌群的丰富度和多样性均显著高于22~28 cm沉积物的数值(P<0.05).变形菌门和绿菌门细菌的相对丰度与沉积物的含水量、总氮和总有机碳含量呈显著正相关关系(P<0.05),芽单胞菌门和放线菌门细菌的相对丰度与沉积物的含水量、总氮和总有机碳含量呈显著负相关关系(P<0.05),厚壁菌门、酸杆菌门、浮霉菌门、硝化螺旋菌门、绿弯菌门和拟杆菌门细菌的相对丰度与沉积物的理化因子间没有显著相关关系(P>0.05).In order to learn the composition and function of wetland microorganisms,rationally utilize and develop wetland microbial resources,16S rRNA(V4-V5 region)of sediment bacteria in Beidagang wetland of Tianjin were examined based on Illumina Mi Seq sequencing platform. The bacterial community structure and diversity at 0-8,8-22 and 22-28 cm sediments were investigated. The results showed that 1 716 OTUs,2 000 OTUs and 1 263 OTUs were identified at 0-8,8-22 and 22-28 cm sediment depth,respectively. Determined categorization of bacteria were 16 phyla,42 classes,70 orders,116 families and 187 genera. Proteobacteria,Chloroflexi and Acidobacteria were the dominant groups at 0-8 cm sediment depth with the relative abundance of 44.73%,16.84% and 10.25% in turn. Proteobacteria(37.24%)and Chloroflexi(14.02%)were the dominant bacterial groups,followed by Acidobacteria(11.27%) at 8-22 cm depth. Firmicutes(45.84%),Proteobacteria(21.95%)and Chloroflexi(6.26%)were the dominant bacterial groups at 22-28 cmdepth. At0-22 cmsedimentdepth,bacterial community richness and diversity were significantly higher than that at 22-28 cm depth(P < 0.05). Pearson′s correlation analysis indicated that the relative abundance of Gemmatimonadetes and Actinobacteria showed significantly negative correla-tions with moisture content,total nitrogen(TN)and total organic carbon(TOC) content(P < 0.05). The relative abundance of Proteobacteria,Chlorobi and Spirochaetae was positively correlated with moisture content,TN and TOC content(P < 0.05).However,the relative abundance of Firmicutes,Acidobacteria,Planctomycetes,Nitrospirae,Chloroflexi and Bacteroidetes did not show significant correlations with soil physicochemical factors(P > 0.05).
