摘  要:目的:观察分析中药益肾疏肝方对去卵巢惊恐大鼠海马Glu/GABA阳性神经元表达的影响。方法:Wistar雌性大鼠60只适应性喂养1周后,随机选取10只行假手术(A组),其余50只去卵巢大鼠随机分为5组:去卵巢组(B组)、模型组(C组)、戊酸雌二醇组(D组)、阿普唑仑组(E组)、中药组(F组),每组各10只。术后第10天起,C、D、E、F组大鼠每日接受"猫吓鼠"1h。同时,各组大鼠连续灌胃28d。末次给药后2h行旷场实验,末次给药后24h逐只取静脉血测定血清E2水平,每组随机选取5只大鼠行心脏灌注,取脑组织,测定海马区Glu、GABA阳性神经元表达。结果:①各组大鼠旷场实验得分:与A组比较,其余四组大鼠水平、垂直活动得分均明显低下,差异有显著统计学意义(P<0.01);与其余四组比较,F组大鼠水平、垂直活动得分均显著升高,差异有显著统计学意义(P<0.01);②各组大鼠血清E2比较:与A组比较,E组E2降低,差异有统计学意义(P<0.05);B、C组E2明显低下,差异有显著统计学意义(P<0.01);F组E2明显高于B、C、E组,差异有显著统计学意义(P<0.01);③海马Glu阳性表达:CA1区:F组显著低于C、D组,差异有显著统计学意义(P<0.01),低于E组,差异有统计学意义(P<0.05)。CA3区:Glu阳性神经元表达:F组显著低于C、D组,差异有显著统计学意义(P<0.01)。海马GABA阳性神经元表达:CA1区:F组显著高于C、D组,差异有显著统计学意义(P<0.01)。CA3区:F组显著高于C、D组,差异有显著统计学差异(P<0.01),高于E组,差异有统计学意义(P<0.05)。结论:中药益肾疏肝方有助于调节去卵巢惊恐大鼠心理异常状态,改善其活动度、探究性;能够参与调节去卵巢惊恐大鼠体内雌激素合成与转化,提高内源性雌激素水平;能够显著降低Glu阳性神经元表达,提高GABA阳性神经元表达,有助于改善惊恐情绪影响下去势大鼠海马CA1、CA3区域的Glu/GABA失衡状态。Objective:To observe and analyze the effect of Yishen Shugan Recipe on the expression of Glu/GABA-positive neurons in the hippocampus of ovariectomized rats.Methods:After 60 Wistar female rats were adaptively fed for 1 week,10 were randomly selected for sham operation (group A),and the remaining 50 ovariectomized rats were randomly divided into 5 groups:ovariectomized group (group B),model group (Group C),estradiol valerate group (group D),alprazolam group (group E),traditional Chinese medicine group (group F),each with 10 rats.From the 10th day after the operation,rats in groups C,D,E,and F received “cat scaring mice” for 1 hour every day.At the same time,rats in each group were given intragastric administration for 28 days.An open field experiment was performed 2 hours after the last administration.Venous blood was taken one by one to determine the serum E2 level 24 hours after the last administration.5 rats in each group were randomly selected for cardiac perfusion,brain tissues were taken,and Glu and GABA positive in the hippocampus were determined.Neuron expression.Results:①The scores of the rats in the open field experiment:Compared with group A,the scores of the other four groups of rats in horizontal and vertical activities were significantly lower,and the difference was statistically significant (P<0.01);compared with the other four groups,F The horizontal and vertical activity scores of rats in the group increased significantly,and the difference was statistically significant (P<0.01);②Comparison of serum E2 of rats in each group:Compared with group A,E2 of group E decreased,and the difference was statistically significant (P<0.05);E2 in groups B and C was significantly lower,and the difference was statistically significant (P<0.01);E2 in group F was significantly higher than that in groups B,C,and E,and the difference was statistically significant (P<0.01);③ Hippocampal Glu positive expression:CA1 area:F group was significantly lower than C and D groups,the difference was statistically signif
