摘  要:目的:观察IL-6干预后人绒毛膜癌JEG-3细胞侵袭能力的变化,并探讨其是否通过STAT3信号通路调控MMPs的表达实现该作用。方法:体外培养JEG-3细胞,实验1分为抑制剂组(5μmol/L STAT3活性抑制剂SD-1008)和空白对照组,处理24 h后,采用Western blot和荧光定量PCR检测2组细胞中p-STAT3蛋白与基质金属蛋白酶(MMP-2、MMP-9)蛋白和mRNA的表达水平。实验2分为IL-6组(10μg/L IL-6)和空白对照组。采用Western blot检测2组细胞处理10、30、60 min p-STAT3的表达水平,采用MTT实验和Transwell实验检测处理36 h后2组细胞的增殖能力和侵袭能力,同时检测MMP-2、MMP-9蛋白和mRNA的表达水平。结果:①与空白对照组相比,抑制剂组的p-STAT3蛋白、MMPs mRNA及蛋白水平均下降(P<0.05)。②与空白对照组相比,IL-6组JEG-3细胞的增殖能力无明显改变,但侵袭能力增强(P<0.05),细胞中p-STAT3、MMP-2和MMP-9的表达量差异无统计学意义(P>0.05),但细胞中活化形式的MMP-2蛋白的表达水平升高(P<0.05)。结论:STAT3可能是调控滋养细胞中金属基质蛋白酶表达信号通路中的上游分子;IL-6可通过上调MMP-2的活化水平来促进JEG-3细胞的侵袭能力,但并非通过STAT3信号通路,其机制有待进一步阐明。Aim:To investigate the changes of invasion ability of choriocarcinoma JEG-3 cells after IL-6 treatment,and further explore whether it functions through STAT3/MMPs pathway.Methods:Human JEG-3 cells were cultured in vitro.In experimentⅠ,JEG-3 cells were assigned to inhibitor group(5μmol/L STAT3 inhibitor SD-1008)and blank control group.After 24 hours,Western blot and qRT-PCR were respectively used to detect the expression of p-STAT3 protein,and the expressions of MMP-2 and MMP-9 protein and mRNA.In experimentⅡ,JEG-3 cells were assigned to IL-6 group(10μg/L IL-6)and blank control group.The expression of p-STAT3 protein in the 2 groups were detected by Western blot after 10,30,and 60 min treatment,the proliferation and invasion abilities of the cells in the 2 groups were detected by MTT and Transwell assay after 36 hours treatment.The expressions of MMP-2 and MMP-9 protein and mRNA were also detected.Results:Compared with blank control group,the expression levels of p-STAT3 protein,MMP-2 and MMP-9 protein and mRNA in the inhibitor group were decreased(P<0.05).Compared with blank control group,the invasion ability of JEG-3 cells in IL-6 group was increased(P<0.05)while the proliferation ability did not change significantly(P>0.05);the expression of activated MMP-2 protein in JEG-3 cells was also increased while those of p-STAT3,MMP-2,and MMP-9 had no significant changes(P>0.05).Conclusion:STAT3 may be the upstream molecule in the signal pathway regulating the expressions of MMPs in trophoblasts,but IL-6 does not regulate the invasion of trophoblasts through this signal pathway,which may up-regulate the invasion of trophoblasts by enhancing the activation of MMP-2.
