黄连等5种中药颗粒剂联合左氧氟沙星对鲍曼不动杆菌生物膜的作用研究

摘　　要：目的:研究黄连、黄岑、五味子、乌梅、连翘等五种中药颗粒剂分别联合左氧氟沙星对多重耐药鲍曼不动杆菌(MDRAB)生物膜的影响。方法:对临床分离的11株MDRAB进行试验,用龙胆紫染色法筛选产膜菌株,再用微量倍比稀释法测定六种药物的最小抑膜浓度,然后五种中药颗粒剂分别与左氧氟沙星用肉汤微量稀释棋盘法进行联合抑膜实验,镜下观察生物膜的变化并记录最小抑膜浓度,最后计算分析FIC结果分布。结果:黄连、黄芩、乌梅、五味子联合左氧氟沙星的FIC表现为协同、相加和无关作用,均以无关作用为主,分别占70%、40%、50%、60%;连翘联合左氧氟沙星的FIC作用结果以拮抗作用为主,占40%。结论:本研究中联合用药的最小抑膜浓度有不同程度差异,除连翘外其余四种中药颗粒剂联合左氧氟沙星表现未见拮抗作用,由于实验数据较少可能导致结果有所偏差,在同类实验中,可考虑选用除连翘外其余四种没有拮抗作用的组合进一步研究。Objective:To study the effects of five Chinese medicine granules including Coptis,Scutellaria,Schisandra,Wumei,and Forsythia in combination with levofloxacin on the biofilm of multidrug-resistant Acinetobacter baumannii (MDRAB).Methods:The 11 strains of MDRAB isolated clinically were tested.The membrane-producing strains were screened by the gentian violet staining method,and then the minimum inhibitory concentration of the six drugs was determined by the microfold dilution method,and then the five traditional Chinese medicine granules were used with levofloxacin.The broth micro-dilution checkerboard method was used to carry out the combined inhibition experiment,observe the changes of the biofilm under the microscope and record the minimum inhibition concentration,and finally calculate and analyze the distribution of the FIC results.Results:The FIC of Coptis,Scutellaria,Wumei,Schisandra combined with levofloxacin showed synergistic,additive and irrelevant effects,all of which were irrelevant effects,accounting for 70%,40%,50%,60% respectively;FIC of Forsythia suspensa combined with levofloxacin The effect is mainly antagonistic,accounting for 40%.Conclusion:In this study,the minimum inhibitory concentration of the combined drugs has different degrees of difference.The other four traditional Chinese medicine granules except Forsythia combined with levofloxacin showed no antagonistic effect.The lack of experimental data may lead to deviations in the results.In similar experiments In addition,the other four combinations that have no antagonistic effect except forsythia can be considered for further study.
